Objective: We can observate myocardial infarct size and adiponectin secretion in rat heart, to discuss the influence of the rosiglitazone(ROS) to the protection of the ischemic preconditioning(IPC).Method: 32 SD male rats were randomly divided into 4 groups by Random and control:â‘ ischemia reperfusion group (I/R group);â‘ ischemic preconditioning + ischemia reperfusion group (IPC+I/R group);â‘¢rosiglitazone + ischemia-reperfusion Group (ROS+I/R group);â‘£rosiglitazone + ischemic preconditioning + ischemia reperfusion group (ROS + IPC+ I/R group). In the I/R group and the IPC + I/R group, to lavage saline 2ml/d orally at 8 am daily 2 weeks before the operation; In the ROS + I/R group and the ROS + IPC + I/R group, to lavage Rosiglitazone 3mg/kg + saline 2ml/d orally at 8 am daily 2 weeks before the operation. Measured the levels of adiponectin by RT-PCR in rat heart, and determined the myocardial infarct size in rat heart with staining method after myocardial ischemia to observed the effects levels of the adiponectin to protective the myocardial ischemia. Result: ROS has a protective effect on ischemic myocardium, ROS + IPC can further enhance myocardial ischemia protection against ischemia. After the experiment we measured, the change of adiponectin levels indicates that: application of ROS + IPC can be increased the expression of adiponectin which can enhance the protective effect for ischemic myocardium.Conclusion: ROS can protect the ischemic myocardium, the ROS + IPC can strengthen the protection of the IPC by increasing the level of adiponectin...
