Partâ… Influence of chronic ethanol intake on the cardiovascular system and hepatic system in ratsObjective: The purpose of this experiment is to study the influence of chronic moderate ethanol administration on the cardiovascular system and hepatic system, in order to answer the question whether moderate ethanol intake could or not protect the cardiovascular system and do harm to the liver.Methods: Fifty male healthy adult SD rats were randomly assigned into five groups(ten rats in each group): control group(sodium chloride, 1ml/kg/d); sucrose group(5% sucrose, 1ml/kg/d); three ethanol groups(low dose ethanol group:50% ethanol, 2.5ml/kg/d; middle dose ethanol group:50% ethanol, 5ml/kg/d; high dose ethanol group: 50% ethanol, 10ml/kg/d). All the rats were treated with intragastric administration for 12 weeks. In the end of the 12th week, the plasma glucose and plasma lipid levels were measured; the heart weight index and the left ventricle weight index were calculated; the tissues from the heart, liver and thoracic aorta were stained with HE.Results: The plasma cholesterol level in sucrose group and three ethanol groups were higher than that of control group (P<0.05 or P<0.01).The triglyceride level in middle dose ethanol group and high dose ethanol group were higher than that of control group and sucrose group (P<0.05 or P<0.01).Blood pressure in middle dose ethanol group and high dose ethanol group were higher than that of control group (P<0.05). With HE staining, slight fatty degeneration in the liver can be found in sucrose group and three ethanol groups, the blood vessel endothelium were not smooth either in these groups. Hepatic cord arranged in disorder in middle dose ethanol group. Connective tissue hyperplasia in the liver portal area and spotty necrosis can be observed in high dose ethanol group. Connective tissue hyperplasia in myocardial interstitium can be observed in sucrose group. Myocardial fiber disarranged in the three ethanol groups.Conclusion: Chronic mild ethanol administration can cause the increase of plasma cholesterol, triglyceride and blood pressure and lead to different degrees of damage in liver, heart and blood vessel, which may do harm to the health.Partâ…¡The feasibility study of metadoxine in prevention of acute ethanol intoxication in miceObjective: To investigate the tolerance function of Male Kunming mice to ethanol and the effects of aldehyde dehydrogenase activator metadoxine on acute ethanol intoxication in the mice.Methods: Male Kunming mice were treated with intragastric administration with five doses of ethanol. The drunkenness rate and mortality rate were calculated, then the tolerance function of the mice to ethanol was evaluated, in order to make the appropriate dose of ethanol selected which was used to establish the animal model of acute ethanol intoxication. The mice were assigned into four groups: saline control group and three metadoxine groups (200mg/kg,500mg/kg,100mg/kg),then observed the effects of metadoxine on the mice model of acute ethanol intoxication and compared the drunkenness rate and mortality rate of four groups.Results: The appropriate ethanol dose to establish the mice model of acute ethanol intoxication was 52% ethanol 0.24ml/10g. Comparing with the saline control group, the drunkenness rate and mortality rate of metadoxine groups increased.Conclusion: Metadoxine can not only prevent acute alcohol intoxication in the mice but also worsen it. The specific mechanism is to be studied.
