Objective:To investigate effects of abnormal estrogen and dyslipidemia on estrogen receptorα,andβ, as well as one of the active ingredients of trifolium pratense is formononetin on the protective effect of the central nervous system, observe the ovariectomized hyperlipidemia rats of APP and ERα,βexpression and mechanisms.Methods:The 40 model was successful in ovariectomized wistar rats were randomly divided into four: the model group, Nilestriol group, low dose of thorns formononetin group, high dose of thorns formononetin group, 10 in each group. Another 10 wistar rats were randomly selected for the sham group. Except the sham group, The remaining four groups application of high-fat diet and sterile surgry, establish the model of ovariectomized and hyperlipidemia. With automatic biochemical analyzer, measure serum concentration of TC, TG, LDL-C, levels. With western-blotting,observe the expression of APP.in hippocampal. And use SYBR GreenⅠPCR to observe estrogen receptorαandβon the level of gene expression.Results:(1) Serum concentration of TC, LDL-C sham operation group is lower than model group P<0.01 , nilestriol group is significantly lower than model group P<0.01. Formononetin high-dose group is also significantly lower than model group P<0.01.(2)Expression level of APP protein in hippocampal of centrol nervus system. model group(0.8534±0.0511) higher than sham operation group (0.3851±0.0288) P<0.05.Both nilestriol group(0.2086±0.0169)and formononetin high-dose group (0.4826±0.0316)are lower than model group(0.8534±0.0511) P<0.05. But the difference between nilestriol group and formononetin high-dose group is statistically significant.(3) gene expression level of APP in hippocampal of centrol nervus system.nilestriol group0.72(0.58-0.89),sham operation group1(0.64-1.56),formononetin high-dose group1.96(1.64-2.35)and formononetin low-dose group are all lower than model group and differences is significant in statistic.(4) gene expression. level of Erαin hippocampal of centrol nervus . model group 1(0.44-2.28)lower than sham operation group 3.97(3.16-4.99),difference is significant in statistic;nilestriol group is 7.94(5.94-9.94)times higher than model group, differences is significant in statistic; formononetin low-dose group is 1.24(114-1.75)times higher than model group differences is significant in statistic; formononetin high-dose group is 2.95(2.10-4.14)times higher than model group, difference is significant in statistic.(5) gene expression.level of ERβin hippocampal of centrol nervus model group is 5.03(3.48-7.26)times lower than sham operation group,difference is significant in statistic, nilestriol group is 16.56(12.38-22.16)times higher than model group, difference is significant in statistic; formononetin low-dose group is1.88(1.71-2.07)times higher than model group, difference is significant in statistic, formononetin high-dose group is 4.99(3.55-7.01)times higher than model group, difference is significant in statistic.(6) Pearson correlated analysis suggested that there has some correlations between mRNA of APP in the in hippocampal of centrol nervus and gene expression. level of ErαmRNA,and the correlated coefficient is 0.97,P<0.05;at the same time, there also has some correlations between mRNA of APP and gene expression. level of ERβmRNA, and the correlated coefficient is -0.833,P<0.05.Though the correlated analysis result we can found that, maybe, formononetinf effects the expression level of APP though selective effects.Conclusion:Estrogen declineing and blood lipids increasing can induce high expression of APP. Formononetin has a role in regulating blood lipids. and affect the expression of ERα,βin model rats hippocampal, but also can repress the high expression of APP in the hippocampus..It provides a theoretical basis in protective effect of the central nervous system.
