Alzheimer's disease (AD) is a kind of neurodegenerative disorders in aged people. It becomes severously economic and social loads for both patients' families and their communities. The patients usually die of AD-associated diseases such as decubital ulcer, bone fracture, pneumonia and so on. The regional distribution prediction of over-60-year-old people by the united nations this year showed that: There are 629 million all over the world (10% of world-wide population ), 134 million in China (10% of Chinese population) in the year of 2002. In the year of 2050, there will be 1964 million all over the world (21% of world-wide population), 437 million in China (30% of Chinese population). The incidence of AD showed increased tendency at present in China. There will be more than 5.5 million AD patients in China in 2050, which is the most among the world-wild countries. In addition, one couple in China must and has to look after at least 4 old people in one family at present and in the future. Therefore, it is very urgent and important for our country's social stability and duration-attainable development to study the mechanisms of developing AD and develop anti-AD drugs.The etiology of AD is not known. About its occurring, there are hypotheses of genetics, estrogen deficiency, chronic aluminum toxication, chronic inflammation,etc. Aluminum has showed a relation to AD pathogenesis. In the brain of AD patients, there is abundant aluminum. Administration of aluminum to animals could induce NFTs, APP protein increase, Abeta sedimentation and neuron loss in both cerebral cortex and hippocampus.According to the theory of traditional Chinese medicine, Shen is thefoundation of life, stores essence, dominates wisdom, masters bones and bears marrow and spine. Marrow and spine produce blood and connect with brain. The etiology of AD is Shen deficiency companying with congestion, sputum stacking and hepatic biotransformational abnormality. Professor Li En suggested that Shen, bone, marrow, blood and brain was a function unit and the study and treatment of senile diseases including AD should be in a unified way as the function unit indicated.In order to go further into the roles of aluminum in AD development and the and-AD mechanisms of TKR, we investigated the effects of chronic Aids intoxication on female rats, studied the preventive effects and mechanisms of TKR on AlCl3-induced rats' AD model, from different tissue atratifications and " unified " point of view.Sixty 6-month-old female SD rats , with body weight ranged from 240g to 260g , were divided randomly into 4 groups named group N, group M, group Tl and group T2, and maintained on rat chow and water ad libitum. For group M, Tl and T2, rat was intraperitoneally injected with 1ml A1C13 solution(9mg/ml) per day, 3-day-injection-and-l-day-not per 4-day, for 18 4-days in all. For group N, rat was intraperitoneally injected with saline instand of the A1C13 solution. Meanwhile, each rat of both group Tl and T2 was stomach-perfused with 1ml TKR solution, 7-day-perfusion-and-l-day-not per 8-day, 2.60g TKR/ml solution for group Tl, 5.2Ig TKR/ml solution for group T2, for 9 8-days in all. Each rat of both group N and M was stomach-perfused with tap water instand of TKR solution. TKR is composed of szechwan lovge rhizome, radix rehmanniae, tuckahoe, Mongolian milkvetch root, rhizoma acori tatarinowii, korean epimedium herb, Chinese angelica, common turmeric , and tuber fleeceflower root.Learning and memory and behavior of rats were tested in Y-electric maze. The soluble protein level in both cerebral cortex and hippocampus was tested with Lowry method. The level of soluble E2, Abeta, and EPO in10cerebral cortex, hippocampus and serum, was measured by radioimmunoassay as kit instruction. The level of both ER a and ER 3 protein in cerebral cortical cells was tested by immunohistochemistry as kit showed.
