Cinefaction-digestion alternate approach of sample processing was proposed in atomic absorption spectrometry, through researching and analyzing the composition of inorgenic elements and amino acids in Mantichorula semenowi. The method was proved acurately and it can shorten the processing time, which was 16 hours shorter than direct cinefaction method. It has low sample blank, little amount of adding acid, few digestion times and easy accessibility. The composition of amino acids was determined by automatic amino acid analyzer and different processing methods, determine conditions were chosen based on different amino acid characters. The result indicated that the beetle contains five essential trace elements, and their mass fractions were 189.7×10-6 in sample; the total mass fractions of proteins were 49.9×10-2 in sample, in which the mass fractions of essential amino acids were 0.501. Moreover, leucine, valine, glycin and glutamic acid were relatively abundant, especially for valine, whose mass fraction was as high as 10.3×10-2 in sample. The sample processing method is innovative in this research on the composition of Mantichorula semenowi and it has valuable significance to sample processing for this kind of biological samples. This research supplied basic information for the composition of Mantichorula semenowi , which provided useful information for the further investigation of its value.
