In recent years,with the changes of diet structure,travel style and life rhythm,overweight rate and obesity rate all show a significant increase in the world.In the same period men's semen quality is on a global downward trend.A large number of studies had shown that the quality of semen in obese patients is abnormal,accompanied by a decline in male fertility.Most obese people had abnormal metabolism of amino acids,vitamins and trace elements.Therefore,exploring the relationship between the levels of seminal plasma amino acids,vitamins and trace elements and the semen quality of obese people is important.This article examined the semen quality of infertile men with different body mass index?BMI?.The concentrations of amino acids,vitamins and trace elements in seminal plasma were determined by high performance liquid chromatography?HPLC?,enzyme linked immunosorbent assay?ELISA?and inductively coupled plasma atomic emission spectrometry?ICP-AES?respectively.Then the correlation analysis was conducted to clarify the relationship between amino acids,vitamins,trace elements and semen quality in obese infertile men.The thesis includes the following three parts:1.Objective to investigate the changes of amino acid levels in seminal plasma of obese infertile men and analyze their relationship with semen parameters.Methods According to BMI,21 subjects were divided into normal group?n=7?,overweight group?n=7?and obesity group?n=7?.CASA semen analysis system was used to analyze the parameters of semen,and the contents of aspartic acid,glutamic acid,histidine,arginine,glycine,threonine,proline,alanine Valine,isoleucine,leucine and phenylalanine in seminal plasma were determined by HPLC.Then analyzed the relationship between these indexes and BMI and semen parameters.Results?1?Compared with the normal group,the sperm concentration and the total number of sperm in overweight group and obesity group decreased significantly?P<0.05?.?2?With the increase of BMI,the aspartate showed a decreasing trend,glycine,eleucine,isoleucine and phenylalanine showed a increasing trend,but there was no significant difference among the three groups?P>0.05?.?The levels of amino acids in seminal plasma and BMI and semen parameters were not related.Conclusion Amino acid levels in obese infertile men have a tendency to change,but their relationship with male fertility remains to be further studied.2.Objective to investigate the changes of vitamins in obese infertile men's seminal plasma and analyze their relationship with semen parameters.Methods 83 subjects were divided into normal group?n=27?,overweight group?n=28?and obesity group?n=28?according to BMI.CASA semen analysis system was used to analyze the parameters of semen,and the levels of folic acid,vitamin B12,vitamin C and vitamin D in seminal plasma were determined by ELISA.Then analyzed the relationship between these indexes and BMI and semen parameters.Results?1?Compared with normal group,semen volume,total sperm count and progressively motile sperm?PR%?in obese group decreased significantly?P<0.05?.?2?Compared with normal group,the level of vitamin B122 in overweight and obese group decreased significantly?P<0.05?.?3?The correlation study found that the levels of vitamin B122 and vitamin D in seminal plasma were negatively correlated with BMI?P<0.05?.The level of vitamin B122 in seminal plasma was positively correlated with the sperm concentration and the total number of sperm?P<0.05?.Conclusion The levels of vitamins in seminal plasma of obese infertile men changed,and the changes of vitamins in seminal plasma may affect the quality of semen.3.Objective to investigate the changes of trace elements in obese infertile men's seminal plasma and analyze their relationship with semen parameters.Methods 124 subjects were divided into normal group?n=39?,overweight group?n=45?and obesity group?n=40?according to BMI.CASA semen analysis system was used to analyze the parameters of semen,and the levels of zinc,copper,selenium,cadmium,lead,iron and manganese in the seminal plasma were determined by ICP-AES.Then analyzed the relationships between the above indicators and BMI and semen parameters.Results?1?Compared with normal group,PR%of overweight group and obesity group decreased significantly?P<0.05?;?2?Compared with the normal group,the levels of Cd,Pb and Zn in the overweight group and obese group were significantly increased?P<0.05?,but the Se level was significantly lower?P<0.05?.?3?The correlation study found that BMI was positively correlated with the levels of Cd,Pb,Fe and Zn?P<0.05?,was negatively related to the level of Se?P<0.05?.Seminal plasma Zn concentration was negatively correlated with PR%?P<0.05?.Seminal plasma Fe concentration was negatively correlated with sperm volume and positively correlated with total number of sperm?P<0.05?.Conclusion The levels of trace elements in the seminal plasma of obese infertile men changed,and the changes may affect the quality of semen.
