Objective To study the effect of Mongolia drug TGLG-1 on the level of serum lipids and antiobesity in hyperlipidemia and obese rats induced by abundant fat food, and to discuss the probable mechanism of adjustive effect, and to screene the functional components of the drug. Method Experiments were executed in two stages. First stage is the empirical study of crude extract,according to body weight, 36 Wistar rats were randomly divided into 4 groups(Control group, Model group, TGLG-1 Treatment group, TGLG-1 Prevention group), there are 9 rats in each group. The rats in the control group were supplied normal food, other rats eat abundant fat food. Before drug administration, blood in the rats was collected to detect the contents of cholesterol (TC), triglyceride (TG), HDL-C(high density lipoprotein-cholesterol) in the serum and judge the model. The rats in the control group and model group were given normal saline solution (1.0ml/100g), TGLG-1 Prevention group were given TGLG-1(1.0ml/100g) from first week to last week, TGLG-1 Treatment group were given TGLG-1(1.0ml/100g) from fifth week to last week. The drug were continuously given 6 weeks, Blood was collected to detect the contents of TG, TC, HDL-C in the serum. Take liver, kidney, and adipose tissue of kidney to compute organ index; to observe the change of liver's morphology of each group and to study the expression of TNF-αin adipose tissue with immunohistochemical method. Second stage is the empirical study of different solvent extraction, according to body weight, 80 Wistar rats were randomly divided into 8 groups, They are Control group, Model group, solvent control group(olive oil), Lovastatin group, ligroin extract group, acetidin extract group, n-butyl alcohol extract group, water extract group, 10 rats for each group. The method of feeding and preparing model are same to first stage. The rats in the control group and model group were given equal-volume normal saline, solvent control group(olive oil) were given isodose olive oil, others were given different drugs. The drug were continuously given 6 weeks, Blood was collected to detect he contents of TG, TC, HDL-C, FFA, AST, ALT, LP in the serum. Take liver, kidney, and adipose tissue of kidney and testis to weight and compute organ index; to observe the change of liver's morphology of each group and study the expression of TNF-αin liver and adipose tissue with immunohistochemical method. Results (1) Changed of lipid content: Compared with the model group, it is found that TGLG-1 crude extract could significantly decrease the level of TC, TG,decrease the level of LDL-C and increase the level of HDL-C in serum of experimental groups. Contrast difference have statistical significance (P<0.01). (2) Changed of weight and organ index: Compared with the model group, TGLG-1 crude extract could significantly decrease weight and organ index. Contrast difference have statistical significance (P<0.01). (3) The result of immunohistochemistry showed that the expression of TNF-αin adipose tissue was lower in TGLG-1 prevention group and treatment group than in the model group, Contrast difference have statistical significance (P<0.01). (4) Compared with the model group, acetidin extractant could decrease the level of TC, TG, FFA, weight and Lee's index (P<0.05), the expression of TNF-a in liver and adipose tissue were lower significantly (P<0.05). (5) The level of leptin in serum in different extractant group have no statistical significance, but the level of AST and ALT in acetidin group were decreased significantly (P<0.05). Conclusions (1) The decrease of weight, adipocyte area, TC, TG, and LDL-C and the increase of HDL-C in hyperlipidemic rats after TGLG-1 administration indicated that TGLG-1 may have the effect of treatment and prevention from hyperlipidemia and obese. (2) The expression of TNF-αwas higher in model group rats than that in TGLG-1 treatment and prevention groups, indicated that TNF-αmay participate the formation of obese and hyperlipidemia induced by abundant fat food. So it is presumed that the mechanism of serum lipids and weight decreasing possibly related to the higher expression of TNF-α. (3) The decrease of weight, Lee's index, and FFA, TC, TG in serum and the lower expression of TNF-αin liver and adipose tissue in hyperlipidemic rats after acetidin extractant administration indicated that the utility component of TGLG-1 may be in acetidin fraction. (4) The level of leptin in serum in different extractant group have no statistical difference, it indicated that the mechanism of serum lipids and weight to be decreased in acetidin group wouldn't have correlation with the adjustment of leptin. (5) The decrease of AST and ALT in acetidin group indicated that the extractant may have the protective effect on hyperlipemia-induced liver injury; it is presumed that it may have the preventive and therapeutic effect for fatty liver.
