| Objective Hypoglycemicâ… originated form Professor La. According to the pathogenesis that cold moves into lung from heart which comes from Inner Canon of Yellow Emperor, La Wanying proposed the indications of Hypoglycemicâ… which marked by upper wasting, polydipsia and polyuria. There have not been related reports about its mechanism on the treatment of diabetes. With years of clinical experience, from the view of the Chinese medicine theory and through the basic experiments and clinical experience professor La Wanying suposes the common pathogenesis of the diabetes is that deficiency of yin fluid, sthenic syndrome of dry and heat. The root cause of the disease is deficiency of yin while the symptom is the syndrome of dry and heat and enduring diabetes will be companied by the blood stasis. The clinical symptoms are polydiasia, polyphagai, polyuria, emaciation and lassitude. Professor La established the diabetes treatment principles of replenishing qi and nourishing yin, promoting fluid production to quench thirst, clearing away heat and promoting blood circulation,modifying the ingredients of the decoction, he also proposed its actions which includes replenishing qi and nourishing yin, promoting fluid production to quench thirst, clearing away heat and promoting blood circulation. Through the experiment observe the influence of Hypoglycemicâ… on the FBG (fast blood glucose), blood-lipid, cholesterol, glycolated hemoglobin, the blood serum insulin, histopathology of pancreas ext. of the type 2 diabetes rat model and discuss the hypoglycemic action. Antiatheroscloresis action and the influence on the development of the type 2 diabetes of the Hypoglycemicâ… on the type 2 diabetes rat model and has a preliminary discussion on its mechanism of action, and provides a theoretical evidence on diabetes treatment of Chinese medicine.Methods Select 70 male Wistar rat, SPF-class, weighted 120±20g and label them with number according to their weight. Select 10 rats to constitute the normal control group randomly and the rest 60 rats constitute the diabetes model group. Feet them with high fat diet and give them enterocoelia injection frequently with small dose of streptozotocin(40mg/kg)to produce type2 diabetes rat molel. Five days later, caudal vein blood collection and check the blood glucose level, and recognise the diabetes model according to the blood glucose level. With the random number table, the model rats are dibided into model control group with 10 rats which are given 0.9 % saline, metformin roup with 9 rats which are given metformin tablets, Ginseng and Asreagalus hypoglycemic particle group with 9 rats which are given Ginseng and Asreagalus hypoglycemic particles, low, medium and high dose of Hypoglycemicâ… group with 9 rats respectively which are given the different doses of Hypoglycemicâ… by intragastric administration. All the rats in different groups are given medicine treatment once daily for 4 weeks respectively and recorded the weight weekly. At the end of the experiment, all the rats are given anesthesia with 0.3 % sodium pentobarbital. Collect abdominal aorta blood and check the level of FBG, TC, TG, HbAlc, Ins and remove and mount the entrail of pancreas for pathological section and immuno-histochemistry. The experiental results are statistically analysized through SPSS13.0 statistical software to compare the changes of FBG, TC, TG, HbAlc, Ins in different groups before and after the treatments separately.Results 3 of 70 rats were removed because their glucose levels don't mach the standar of model rats. 2 of 70 rats were dead due to the overhigh glucose level during the modeling. 65 rats survived to the end of the experiment and divided into Nornal group with 10 rats, Model control group with 10 rats, Metformin group with 9 rats, Ginseng and Asreagalus hypoglycemic particle group with 9 rats, and low, medium and high dose of Hypoglycemicâ… group with 9 rats respectively sum up to 27 rats.1 weight The weight of rats in each group has no significant difference before treatment (P>0.05); the weight of the rats in Modeling group decreased significantly after 4 weeks'high-fat diet and intraperitoneal injection of STZ (P<0.05); the weight of the rats in the Modeling group has no significantly different (P>0.05); the weight of rats in medicine-given group and Modeling group were significantly different (P<0.05).2 blood glucoseCompared with the normal group, the level of blood glucose in medicine-given group and medeling group decreased significantly after 4 weeks'treatment(P<0.05);the effect of low dose of Hypoglycemicâ… is more effective than that of medium and high-dose group which is significant (P<0.05); the effect of low dose of Hypoglycemicâ… has no statistical significant difference with that of the metformin group and Ginseng and Asreagalus hypoglycemic particle group(P>0.05).3 Hemoglobin-AlcCompared with the rats of model group, the content of Hemoglobin-Alc in medicine-given groups'rats were decreased dramatically (P<0.01).4 LipidsCompared with the rats of model group, the content of TC, TG in medicine-given groups'rats were decreased dramatically(P<0.05).5 InsulinCompared with the rats of model group, Hypoglycemicâ… can increase the content of serum insulin of T2DM model and have significantly protective effect on the injured pancreatic.6 Observation of Histopathological of pancreasCompared with the rats of model group, the isletβ-cell number in medicine-given groups'rats were increased whose morpha were relatively regular, structure relatively normal, nucleolus relatively clearer and cells arrangement regular. Compared with the rats of model group, the isletβ-cell number in Hypoglycemicâ… groups'rats were increased significantly whose cells distribution regular, necleolus clear, cells arrangement regular, nuclear size basically equality.7 Observation under the Immunohistochemical light microscopyCompared with the rats of model control group, the isletβ-cell number in medicine-given groups'rats wererelatively increased. Compared with the rats of model control group, the isletβ-cell number in Hypoglycemicâ… groups'rats were increased significantly whose morpha were relatively regular, structure relatively normal.Conclusions1 Hypoglycemicâ… can decrease the fasting blood glucose level of T2DM rats.2 Hypoglycemicâ… can umprove lipid metabolism of T2DM rats.3 Hypoglycemicâ… can reduce the content of glycated hemoglobin of T2DM rats.4 Hypoglycemicâ… can increase the serum insulin level of T2DM rats.5 Hypoglycemicâ… has better therapeutic effect on the T2DM rats which promotes pancreatic cells repair.6 Rat-model of type 2 diabetes can be established through continuously giving high-fat diet and multiple intraperitoneal injections of low dose of streptozotocin. |
