Different Doses Of Streptozotocin Induced Type I Diabetes Animal Model In Rhesus

Objetive To evaluate the efficiency and side effect of different dose of STZ to diabetic rhesus monkeys,search for the optimal dose of inducing type 1 diabetes models and provide safe and stable animal model for relative diabetic investigation.Methods We select 15 mature,health,male rhesus monkeys, randomly divided into three groups,among the total,there are 3 in the high-dose group 100mg/kg,4 in the middle-dose group 75mg/kg,8 in the low-dose group 50mg/kg,intravenous injection certain dose of streptozotocin,testing the blood glucose,C peptide,intravenous glucose tolerance test(IVGTT),liver and renal function and the weight change and so on regularly,do pathological examination of hepar,renal,pancreas tissues.Results The animals were died in two weeks when injected 100mg/kg STZ once.When injected 75mg/kg STZ,the animals all developed diabetes,the AST,ALT,UREA values all increased significantly when tested 1,2 weeks after building models,varying degrees steatosis were found in hepar histopathological examination,2 in 4 were died in 2 months after building models.The success rate was 75% when injecting 50mg/kg STZ and no animal died during experiment,no obvious abnormality was found in liver and kidney functions and pathological examination,C peptide was non-response to glucose stimulation in IVGTT more than 1 year during maintenance of diabetes status.All the 3 groups were polydipsia,polyuria,more food and lossing weight.The amount ofβcells of islet were decreased significantly in pancrease immunohistochemical staining.Conclusion The dose of 75mg/kg STZ has the highest success rate,but has the bad impact on liver and renal function and the animals have short survival time,but the 50mg/kg STZ group can extend the duration of animal models,maintain a high level of blood glucose status for one year which do favor to our next study though the success rate of animal model was lower than the middle-dose group.
