PURPOSE: To investigate the differences of B7-H3 and TLT-2 expression between in oral squamous cell carcinoma tissues and normal oral mucosa, then further research the function and significance of B7-H3 expression on the biology of oral squamous cell carcinoma cells.METHODS: B7-H3 and TLT-2 expression in oral squamous cell carcinoma tissues and normal oral mucosa were detected by RT-PCR/RT-qPCR and immunohistochemical assay. B7-H3 adenovirus expression vector was constructed, and Tca8113 cells were transfected. Then we have used fluorescence microscope to observe GFP fluorescence intensity and density, in order to detect infection efficiency of Adv-pEGFP-B7-H3. B7-H3mRNA expression in each group cells were detected by RT-qPCR, then the cell proliferation and cell cycle were evaluated by CCK-8 and flow cytometry (PI staining) assay, respectively. The date was analyzed with SPSS 11.0 software package.RESULTS: RT-PCR results showed B7-H3 and TLT-2mRNA are both expressed in oral squamous cell carcinoma and normal oral mucosa. But RT-qPCR results showed that purpose gene have difference expression between two groups. In oral squamous cell carcinoma, the expression of B7-H3mRNA was 7.2904±0.3647 times significantly higher than normal oral mucosa, and the expression of TLT-2mRNA was 5.8573±0.3504 times significantly higher than normal oral mucosa. After Tca8113 cells were transfected by B7-H3 adenovirus expression vector for 24h, compare with the control group, cell proliferation and S-phase fraction of 10, 50, 100 MOI group increased significantly (P<0.05); At the same time, compared with 1 MOI group, cell proliferation and S-phase fraction of 50, 100 MOI groups increased significantly (P<0.05), however, significant difference was not observed between 50 and 100 MOI group (P>0.05).CONCLUSION: In oral squamous cell carcinoma tissues, B7-H3/TLT-2 mRNA and protein expression were significantly higher than normal oral mucosa; in vitro, B7-H3 high expression can promote the proliferation of oral squamous cell carcinoma cells.
