| Purpose: By transmission electron microscopy and deoxyribose nucleotide end transferase mediated gap end labeling (TUNEL) method used to observe the BuShenZhuangGu capsule capsule on experimental rabbit knee OA cartilage cell apoptosis influence and RT-PCR detect Bax Bcl-2 in mRNA level express changes to investigate the experimental rabbit knee OA mechanism and clinical curative effectMethods: Experimental research, 40 male New Zealand big white rabbits were bred one week to adapt the environment.Then they were randomly divided into four groups: normal group, model group, the treatment group and control group.We manufactured rabbit knee OA model by improved Hulth method.Model group, the treatment group and control group did unilateral knee anterior cruciate ligament QieDuanShu (aclt).The treatment group was gived the BuShenZhuangGu capsule,while the control group was gived ZhuangGuGuanJie pills.The knee is filming for CR after 8 weeks, after the rabbit executed by using TUNEL method used to observe the BuShenZhuangGu capsule on experimental rabbit knee OA cartilage cell apoptosis influence, RT -PCR detect Bax, Bcl-2 in mRNA level express changes.Clinical research BuShenZhuangGu capsule and ZhuangGuGuanJie pill were each used to therapy 30 patients, through the symptoms and signs of improvement observed the clinical curative effect of two drugs.Results: The experimental research shows that in normal group of apoptosis index is smallest,and the model group of apoptosis index is highest, two groups of significant difference (P < 0.05), the treatment group and control group apoptosis index obviously less than model group with significant difference (P < 0.05), the treatment group and control group apoptosis index are higher than normal group, a statistically significant (P < 0.05). Can conclude, BuShenZhuangGu capsule can inhibit the Bax , promoting Bcl-2 expression, improved Bcl-2 / Bax ratio.Bax/Bcl-2 ratio in model group was obviously higher than that of normal, treatment group and control group and model group compared ratio drops, difference was statistically significant (P < 0.05). Treatment group and control group comparison, ratio no difference obvious (P > 0.05). Clinical studies showed that application of the BuShenZhuangGu capsule for knee OA total effective rate is over 94%.Conclusion: Bcl-2 family through the protein and protein interaction regulating cell survival and apoptosis. Among them, inhibiting cell apoptosis Bcl-2 and promoting cell apoptosis Bax are regulating center. The BuShenZhuangGu capsule can inhibit the Bax capsule, promote Bcl-2 expression, improved Bcl-2 / Bax ratio, inhibit joint cartilage cell apoptosis.
