Objective To investigate the effect of triptolide and hydrocortisone on inflammation factor IL-1β, TNF-αexpression by macrophage ,and explore the therapy mechanism of triptolide and hydrocortisone on inflammation.Method 1,The peritoneal macrophages of mice were separated, purifed and activated by LPS in this study, the were cultured in vitro with a series concentration of TP and HC during different time.2, The concentrations of IL-1βwere measured by ELISA.3, The activities of TNF-αwere measured by MTT colorimetry.Result 1,The levels of IL-1βand TNF-αproduction were significantly inhibited by triptolide(0.1~10ug/ml).2, The levels of IL-1βand TNF-αproduction were significantly inhibited by hydrocortisone (20~80ug/ml).Conclusion Triptolide and hydrocortisone inhibits IL-1βand TNF-αproduction by macrophage suggesting that the inhibitory influence of triptolide and hydrocortisone on IL-1βand TNF-αproduction may be one of the mechanisms underlying its therapy effect on inflammation.
