Objective To investigate the effect of triptolide on vascular endothelial growth factor (VEGF),TNF-αand IL-8 expression by macrophage, and explore the therapy mechanism of triptolide on endometriosis.Method 1. The peritoneal macrophages of mice were separated ,purified and activated by LPS in this study, then were cultured in vitro with a series concentration of TP during different time. 2. The levels of VEGF mRNA,TNF-αmRNA and IL-8 mRNA expression were measured by RT-PCR. 3. The effects of triptolide on VEGF production were measured by immunohistochemical method. 4. The activities of TNF-αwere measured by MTT colorimetry. 5. The concentrations of IL-8 were measured by ELISA.Result 1. The levels of VEGF,TNF-αand IL-8 production were significantly inhibited by triptolide (1~10ug/ml). 2. The levels of VEGF mRNA,TNF-αmRNA and IL-8 mRNA expression were significantly inhibited by triptolide(1~10ug/ml) . Conclusion Triptolide inhibits VEGF,TNF-αand IL-8 expression and production by macrophage, suggesting that the inhibitory influence of triptolide on VEGF,TNF-αand IL-8 expression and production may be one of the mechanisms underlying its therapy effect on endometriosis.
