Objective To study the expression changes of claudin-4 in the lung tissues of rats with acute hemorrhagic necrotizing pancreatitis and its significances.Methods In accordance with the completely random way, the 42 Sprague-Dawley rats were divided into AHNP group(n=21) or Sham-operated group(n=21), 21 rats in each group. Animal models with acute hemorrhagic necrotizing pancreatitis were induced by injection of 5% sodium de-oxycholate solution through common biliopancreatic ducts. Sham group rat was opened the abdominal cavity without injection, and then the abdomen was closed. Animals were sacrificed at 12 hours later. Pancreas and lung tissues were harvested for the histopathological examinations. The wet weight and dry weight of the right lung of the rats was detected for the wet/dry ratio. The claudin-4 protein in the lung tissues of rats were analyzed by the method of Western blotting.Results There were significant pathological changes in both pancreas and lung tissues of the acute hemorrhagic necrotizing pancreatitis rats. Compared with sham-operated group, lung wet/dry weight ratio of rats in AHNP group was significantly higher (P <0.01) and the claudin-4 protein expression of lung tissue of rat in AHNP group was significantly lower (P <0.05).Conclusion There is obvious lung injury of rats with acute hemorrhagic necrotizing pancreatitis. The expressions of claudin-4 in the lung tissue of the rats with acute hemorrhagic necrotizing pancreatitis decrease significantly. And this may play an important role in acute lung injury.
