Objective: To study the expression and clinical significance of tumor metastasis suppressor gene KAI1 and MMP-2 in breast cancer.Methods: The expression of KAI1 and MMP-2 were detected by immunohistochemistry in 75 specimens of breast cancer tissue and 14 specimens of benign breast diseases tissue and its relationship with pathological type, clinical stage, histological grade, metastasis of axillary lymph nodes, tumor size, patients' age and menopause were analyzed.Results (1) The positive expression rate of KAI1 in 75 cases of breast cancer was 46.7%(35/75), respectively, which was significantly lower than in benign breast diseases(78.6%)(P<0.05). The positive rate of them had significantly difference. The positive rate was 34.8%(16/46) in the group with metastasis of axillary lymph nodes and it was 65.5%(19/29) in the group without metastasis on the other hand. The positive rate of them had significantly difference. The positive rate was 73.3%(11/15) in the group of intraductal carcinoma and it was 40%(24/60) in the group of infiltrating ductal carcinoma. With the clinical stage and histological grade increasing, the expression of KAI1 in breast cancer decreased gradually(P<0.05). The expression of KAI1 was not associated with tumor size,menopause and the patients' age, but associated with clinical stage, histological grade, metastasis of axillary lymph node and pathological type. (2) The positive expression rate of MMP-2 in 75 cases of breast cancer was 64%(48/75), which was significantly higher than in benign breast diseases (28.6%). The positive of rate MMP-2 was 73.9%(34/46) in the group with metastasis of axillary lymph nodes and it was 48.3%(14/29) in the group without metastasis on the other hand. The positive rate of them had significantly difference. The positive rate was 33.3%(5/15) in the group of intraductal carcinoma and it was 71.1%(43/60) in the group of infiltrating ductal carcinoma. With the clinical stage and histological grade increasing, the expression of MMP-2 in breast cancer increased gradually(P<0.05). The expression of MMP-2 was not associated with menopause and the patients' age, but associated with clinical stage, histological grade, metastasis of axillary lymph node,pathological type and tumor size. (3)The expression of KAI1 was significantly negatively correlated with expression of MMP-2(P<0.001) in breast cancer.Conclusion (1) The expression of KAI1 and MMP-2 were associated with clinical stage, histological grade, metastasis of axillary lymph node and pathological type. Decreased expression of KAI1 and increased expression of MMP-2 promote the invasiveness and metastasis of breast cancer. (2) The expression of KAI1 was significantly negatively correlated with expression of MMP-2 in breast cancer, it suggest that they may participate in progression of breast cancer and have relationship with each other. Combined detect of KAI1 and MMP-2 may have some value for evaluating the potential of invasiveness and metastasis and judging prognosis of breast cancer.
