ObjectivesTo investigate the effect of trehalose in rats evaporative dry eye and its therapeutic mechanisms.MethodsFifty-five SD rats were randomly divided into three groups:normal group5(10 eyes), control group25(50eyes), experimental group25(50eyes) .The normal group wasn't given any disposal,the latter two groups were mechanically prevented from blinking and exposed both cornea and conjunctiva 24 hours to produce vaporative dry eye model.Starting with the model were given sterile saline water,100mmol/L trehalose eyedrops seperately,4 times per day, 2 weeks along.At modeling Instant,1th day,3th day,7th day,14th day of testing SchirmerⅠtest ,Tear film break-up time, fluorescence staining.After inspection,taking the cornea and conjunctiva tissue for HE and Immunohistochemical staining,taking pathological examination and observing expression of cytokines-- TNF-α,IL-6.Results1.SⅠt results comparison: there were no significant statistical differences through three groups(P>0.05).2.BUT results comparison:no significant differences in normal group between every two observing points(P>0.05);there was statistical significance between control and experimental group on 7th day and 14th day(P<0.05);14th day between normal and experimental group showed statistical significance(P<0.05).3.FLS results comparison:fluorescence staining points were 0 in normal group(P>0.05);Staining points reach the maximum at the time as modeling in both control and experimental groups,and on each observing time in control group it showed no significant difference(P>0.05);the difference became statistical between experimental and control group since 3th day(P<0.01).4.Immunohistochemical staining comparison:A large amount of TNF-α,IL-6 expressed in the latter two groups after modeling.the mount deduced in the control group,obviously deduced in the experiment group.There were statistical differences on 14th day between every two groups(P<0.01).ConclusionsTrehalose can inhibit the expression of inflammatory cytokines TNF-α,IL-6 on the surface of corneal and conjunctiva,and relief inflammatory reaction on corneal and conjunctiva surface.It was suggested that there were prospects in the therapy of dry eye by trehalose.
