| ObjectiveTo investigate the significance in expression of 14-3-3σand CyclinB1 in infiltrating breast carcinoma as well as the relationship in prognosis.MethodsThe expression of 14-3-3σand CyclinB1 protein were detected with PV-9000 immunohistochemistry (IHC) and western blot analysis in infiltrating breast carcinoma, breast ductal carcinoma in situ, atypical ductal hyperplasia, breast usual ductal hyperplasia as well as breast adjacent carcinoma tissues.Results(1) Immunohistochemistry results showed that the positive expression rate of 14-3-3σprotein was 22.5% (18/80) in infiltrating breast carcinoma, significantly lower than 65%(26/40) of breast ductal carcinoma in situ,86.7%(13/15) of atypical ductal hyperplasia,92%(23/25) of breast usual ductal hyperplasia and 96%(24/25) of breast adjacent carcinoma tissues (P<0.05); 14-3-3σprotein expression in infiltrating breast carcinoma was relevant to histological grade,TNM stage and axillary lymph node metastasis (P<0.05), however, irrelevant to age and the tumor size (P>0.05).The expression quantities of 14-3-3σdetected by western blot were consistent with the results of IHC. (2) The positive expression rate of CyclinB1 protein was 67.5%(54/80) in infiltrating breast carcinoma, significantly higher than 52.5%(21/40) of breast ductal carcinoma in situ,46.7%(7/15) of atypical ductal hyperplasia,36%(9/25) of breast usual ductal hyperplasia and 28%(7/25) of breast adjacent carcinoma tissues (P<0.05); CyclinB1 protein expression in infiltrating breast carcinoma was relevant to axillary lymph node metastasis (P<0.05). (3) 14-3-3σand CyclinB1 protein expression in infiltrating breast carcinoma was negatively correlated (rs =-0.521, P=0.000). (4) The positive expression rate of 14-3-3σin infiltrating breast carcinoma with postoperative recurrence group 5.9%(2/34), significantly lower than without recurrence group 39%(16/41) (P<0.05); but the positive expression rate of CyclinB1 in infiltrating breast carcinoma with postoperative recurrence group 97.1%(33/34), significantly higher than without recurrence group 51.2%(21/41) (P<0.05). (5) Retrospective analysis the survival time of patients with infiltrating breast carcinoma showed that 14-3-3σprotein positive expression group patients of the 1, 3, 5-year survival rate insignificantly higher than the negative expression group, OS, median TTP, MST insignificantly longer than the negative expression group (P>0.05); CyclinB1 protein negative expression group patients of the 1, 3, 5-year survival rate insignificantly higher than the positive expression group, OS, median TTP, MST insignificantly longer than the positive expression group (P>0.05)。Conclusions(1) In infiltrating breast carcinoma ,14-3-3σprotein expressed low, related to histological grade,TNM stage,axillary lymph node metastasis and postoperative recurrence, CyclinB1 protein expressed high, related to axillary lymph node metastasis and postoperative recurrence ,suggesting that it may play an important role in infiltrating breast carcinoma carcinogenesis and development.(2) 14-3-3σand CyclinB1 protein may all cannot become independent factors to evaluate the prognosis of infiltrating breast carcinoma. |
