Objective:Study on Technology of Extraction of Polysaccharide in Saccharum Alhagi and Separation,Purification Polysaccharide in Saccharum Alhagi. Study on polysaccharides in Saccharum Alhagi in nature and sugar formed,Antioxidant Antioxidant activity and preliminary analysis the separated compounds of the structure.To establish the spectrofluorimetry of determining amino acid in saccharum alhagi.Methods:1. Determined best environment of polysaccharides in Saccharum Alhagi by the Orthogonal test.2.Adopt DEAE-52 Fiber and Sephadex G-150 Separate and Purificate polysaccharides in Saccharum Alhagi.3. Research Rationalized nature of S1 and Adopt TLC analysis Monosaccharide composition.4.ThroughUV,IR,NMRanalysis Components of polysaccharides.5.Study on the Antioxidant activity of Polysaccharide in Saccharum Alhagi.6.Amino acid was extracted from saccharum alhagi using distilled water by the way of ultrasound, and deriveed by 0.1% OPA, then determined by spectrofluorimetry. Results:The best technology is 70℃,nian times water,Have extracted 1.5h, concentration of the alcohol for precipitate is 80 percent..2.Adopt DEAE-52 Fiber and Sephadex G-150 Separated,obtain six components.3. Preliminary assessment Monosacc -haride composition is Glucose.4. The Result of Structural analysis are D-Glu-1α→2β-D-Fru and D-Glu-1β→6-D-Glu-1α-2β-D-Fru.5. Antioxidant activity of Polysaccharide in Saccharum Alhagi make clear S1 is a good Antioxidants.6. The linear range was 3.0952~15.5071μmol·L-1(r=0.9990). The limit detection was 9.70×10-7 mol·L-1. The average recovery of samples was 93.6-98.9%(RSD=2.5%). Conclusion: 1.Pass the test, Verify results is Similar as the result of orthogonal test,this account for the technics is steady and practicable.2. Through DEAE-52 Fiber basic achieved effects.3.Initial judged Monosaccharide composition is Glucose.5. polysaccharide's action of removing Radicals and Antioxidant activity could stop Radical's oxidative damage.6. The method is simple, rapid and high sensitivity, which can be used for the determination of total free amino acids in natural drugs.
