Objective:To research and assess bentonite on the protection of gastric mucosal injury in order to preparate aspirin slice tablet of easing gastric mucosal injury. Methods:①Gastric mucosal injury was induced by aspirin, while ulcer index(UI)was observed and calculated, do gastric histopathology, gastric mucosal MDA content and SOD, GSH-Px activity were measured in serum, pH value of stomach contents were assayed, Gastric barrier mucus was quantitatively determined by means of Alcian blue to stain acidic mucus.②Using the single factor experiment design, based on the external appearance, hardness, crumbling time, to sieve the molding process.The quantitative determination of aspirin was performed with HPLC method.③From appearance of tablet, hardness, brittle broken degree and dissloution curve these aspects, compare the original aspirin slice with the new ones.④Gastric mucosal injury was induced by aspirin, ulcer index(UI)was observed and calculated, do gastric histopathology.⑤The experiments were carried out in rabbits and Old prescription aspirin plates were used as contro 1.The main parameters of phamaeokinetics were calculated by DAS PK software. In vitro and in vivo tests were developed and compared with the control tablets.Results:①The pathological examination and biochemical tests found that when bentonite dose was 50 mg·kg-1, the gastric mucosa of the mice had a protective effect, when the bentonite dose was 200 mg·kg-1~300 mg·kg-1, it can significantly improve gastric mucosal damage, inhibit MDA of gastric mucosal, restore the activity of SOD and GSH-Px, and in this context gastric mucus content also significantly increased, but the pH value of stomach contents had no significant effect.②The best recipe was aspirin 0.1g, bentonite 0.25g,0.001g folic acid, 0.4% micro powder silica gel. The standard curve was linear over the concentration range of 0.1~1mg·mL-1 for aspirin. The recovery of aspirin was 99.25% with RSD 0.4%.③The two sides from tablet appearance, hardness, brittle broken degree were almost no difference, dissloution curve was similar.④Old prescription group had obvious ulcers of mice gastric mucosa,new prescription group did not see ulcerative, had significant differences.⑤Bioequivalence of new prescription aspirin tablets is consistent with the old one.Conclusion:New prescription aspirin tablets can alleviate stomach mucosa damage.
