| Objective:To investigate the effect of dynamic follow-up minimal residual disease (MRD) of leukemia patients by fluorescence in situ hybridization (FISH),assess the treatment by MRD and predict the occurrence of GVHD. Methods:85 specimens from 44 leukemia patients after chemotherapy or allogeneic stem cell transplantation were studied by FISH and conventional cytogenetic analysis. Results:1.FISH analysis showed that 15(17.6%) samples were positive. Conventional cytogenetic analysis showed that 11(12.9%)samples were positive. The two methods have no significant difference;2. MRD-positive patients recovered after the appropriate treatment. The FISH results after transplantation have no difference in statistics (P>0.05);3.The result of FISH showed that the percentage of positive cells in the treatment group were lower than the before treatment group (P<0.01), but still can not believe that the drug treatment group and the hematopoietic stem cells transplant group have difference in statistics (P>0.01); 4. The incidence of acute graft-versus-host disease (GVHD) has no correlation with the results of FISH of 1 month after transplantation (P>0.05). Conclusion:1. FISH and conventional chromosome analysis has no significant difference in detection rate, but faster, more sensitive, more suitable for clinical use;2.Using FISH monitoring MRD is important to evaluate the status of patients and guide corresponding treatment;3.FISH can reflect the treatment effect accurately;4.MRD still can not accurately predict the occurrence of GVHD. |
