Objective:To investigate the effects of total phenolics from abnormal savda munziq combined with cisplatin or docetaxel on Hela cell line of cervical cancer cultured in vitro, to look for better treatment to cervical cancer. Methods:Hela cells were cultured in vitro and treated with different drug concentrations;the cisplatins, the docetaxels, the total phenolics, the total phenolics combined with cisplatins and the total phenolics combined with docetaxels. Microscope was used to observe the growth of Hela cells. The inhibiting effects were tested by MTT assay, and flow cytometry was used to examine cell apoptosis rate, Bcl-2 and telomerase, real time-PCR was used to detect FHIT and Survivin's expressions. Results:1. The inhibiting effects of total phenolics, cisplatin and docetaxel were obvious, the combinations were moer obviously, the differences had statistical significances (P<0.05). The low dose of total phenolics can't reduce the inhibiting effect of Hela cells, but it can increase the cisplatin and docetaxel's inhibiting effect. Hela cells showed Morphologic changes obviously in experimental groups.2. The 48h's apoptosis rates of total phenolics, cisplatin and docetaxel increased (P<0.001), they were (12.80±0.70)%, (18.87±3.79)%and(15.87±3.81)%;the combinations had interactions (P<0.01), the rates were (31.87±3.82)%and (36.47±3.98)%.3. The 48h's Bcl-2 rates of total phenolics, cisplatin and docetaxel were reduced obviously (P<0.001), they were (63.63±3.84)%, (49.20±4.08)%, (51.40±6.35)%;the total phenolics combined with cisplatin had interactions (P=0.019), the rates were (14.57±2.32)%;the total phenolics combined with docetaxel had no interactions, but it still induced Bcl-2 rate; (18.93±4.83) %, there were differences between groups (P<0.001).4. The 48h's telomerase rates of total phenolics, cisplatin and docetaxel were reduced obviously (P<0.001), they were (51.37±4.68)%, (40.83±5.30)%, (36.83±5.26)%;the combinations had no interactions, but they still decreased Bcl-2 rate; (14.90±2.50)%,(18.93±3.97)%, there were differences between groups (P<0.001).5. The 48h's FHIT expressions of total phenolics, cisplatin and docetaxel increased obviously (P<0.001), the combinations had interactions (P<0.001).6. The 48h's Survivin expressions of total phenolics, cisplatin and docetaxel increased obviously (P<0.001), the combinations had interactions (P<0.001) Conclusion:The total phenolics from abnormal savda munziq can inhibit Hela cells, and it can improve cisplatin and docetaxel's inhibition, the combinations have greater inhibition. When cisplatin and docetaxel's concentrations are definite, total phenolics from abnormal savda munziq has in time-indepent and dose-indepent manner. Total phenolics from abnormal savda munziq combined with cisplatin or docetaxel can decrease Hela cell's apoptosis obviously, and its mechanism may have relations with decreasing Bcl-2, telomerase, Survivin and increasing FHIT.
