Objective:To explore the effects and mechanisms of total phenolics from abnormal savda munziq on SiHa cell line, then to propose the possible treatment of total phenolics from abnormal savda munziq on human cervical carcinoma in theory. Methods:1) MTT assay was used to determine the growth inhibition ratio and IC50 on SiHa cells. Microscope was used to observe the growth of SiHa cells.2) Flow cytometery (FCM) was used to detect the effects of apoptosis rates, Bcl-2 and telomerase.3)Real-time PCR was used to examine the expressions of FHIT and Survivin. Results:1) Total phenolics inhibited the proliferation of SiHa cells in the concentrations from 75μg/ml to 175μg/ml; the inhibition was in dose-effect and time-effect relationships (P<0.01).2) The apoptosis rates were in dose-effect and time-effect relationships, and SiHa cells showed Morphologic changes obviously after total phenolics.3) Total phenolics could decrease the expressions of Bcl-2 and telomerase, and the effects were in dose-dependent manner (P<0.05).4) Total phenolics could decrease the expression of Survivin and increase the expression of FHIT (P<0.05). Conclusion:1) Total phenolics inhibit the growth of SiHa cells in dose and time dependent manners.2) Total phenolics induce the apoptosis of SiHa cells, and its mechanisms may have relations with decreasing the expressions of Bcl-2, Survivin, telomerase and increasing the expression of FHIT.
