Object To investigate the effect of smoking on the expression of Cytochrome P450 2A6 in human bronchial mucosa, and analyse the influence of gender, age and intravenous cephalosporins and inhalation ipratropium bromide and ambroxol hydrochloride on CYP2A6 mRNAMethod 26 cases who underwent pneumonectomy in the Second Xiangya hospital of Central South university chest surgery department were included in this study. All of them were divided into 2 groups: non-smokers and smokers. Besides, we divided smokers into smoking index<800 group and smoking index≥800 group. Using real-time fluorescent quantitative PCR method, the expression of CYP2A6 mRNA was observed in surgically removed bronchial mucosa from all the patients. The comparative CT method (2-ΔCt) was used to calculate the relative quantification of gene expression. All statistical analyses were done by SPSS 17.0 software package. P<0.05 was considered statistically significant.Results All of the cases were detected the expression of CYP2A6 mRNA in bronchial mucosa. There were no relationship between CYP2A6 mRNA and factors of age and inhalation ipratropium bromide and ambroxol hydrochloride. Gender and intravenous cephalosporins were related to CYP2A6 mRNA, but linear regression analysis showed no statistically significant (P>0.05). Smoking index in the smokers was 781.33±330.06, the expression lever of CYP2A6 mRNA in the smokers (2-ΔCt×10000:11.38±6.76) was lower than that in the non-smokers (2-ΔCt×10000:19.87±10.92) (standard regression coefficientβ=-0.444, t=-2.426, P=0.023). The CYP2A6 expression in smoking index<800 group and smoking index≥800 group were 7.29±2.7 and 13.64±7.36 respectively, and there was statistical difference (t= 2.310,P=0.041).Conclusion (1) The smokers have lower CYP2A6 mRNA lever in bronchial mucosa compared to the non-smokers, and smoking may down-regulate the the expression of CYP2A6 in bronchial mucosa. (2) The heavy smokers may have higher CYP2A6 mRNA lever. (3) The expression of CYP2A6 may not influenced by the factors of gender, age, intravenous cephalosporins and inhalation ipratropium bromide and ambroxol hydrochloride.
