Objective: The present study was undertaken to detect the association of PCSK9 E670G polymorphism with coronary heart disease and serum lipid level.Clarify the association of serum PCSK9 level with coronary heart disease and serum lipid level in the Hengyang Han populations in Hunan.Methods: A total of 100 control group and 100 participants of coronary heart disease were randomly selected from our hospital.The levels of TC,TG,HDL-C,and LDL-C were measured using CHOD-PAP,GPO-PAP,Cholestest LDL,Cholestest N HDL respectively.Genotypes of the PCSK9 E670G polymorphism were determined via polymerase chain reaction and restriction fragment length polymorphism combined with gel electrophoresis and then confirmed by direct sequencing. The serum PCSK9 level was measured using ELISA.Results: As to the know SNP-23968A>G,PCSK9 gene allele frequencies of A,G were 95.1% and 4.9% in control group and 86.8% and 13.2% in CAD group, respectively.There was significant differences of alleles and genotypes between CAD group and control group(P<0.05).However,in CAD Patients there was significant difference in TG,TC,LDL-C levels between AA and GG genotype(P<0.05),and there was significant difference in TG between AA and AG genotype(P<0.05).In CAD Patients there was significant difference in the number of diseased vessels of coronary artery and prognosis between AG and AA genotype(p<0.05).There was no significant difference of serum NARC-1 level between the control group and CAD group.Conclusion:1. There was significant differences of PCSK9 alleles and genotypes between CAD group and control group in Hunan Han population. 2. The PCSK9 E670G polymorphism can significantly increase Plasma lipoprotein cholesterol.3. PCSK9 E670G polymorphism had a close relationship with the severity of coronary artery Lesions.4. Plasma PCSK9 levels have not been used as new serum marker of coronary heart disease.
