Effects Of β-elemene On Invasion And Metastasis Of Bladder Cancer T24 Cell Lines

Objective: To explore the effects ofβ-elemene on invasion and metastasis of the Bladder Cancer T24 Cell lines and to investigate the possible mechanism.Methods:Bladder Cance T24 cell lines were cultured in vitro and were treated with 0.01,0.02,0.03mg/Lβ-elemene for 24 h respectively.The samples were detected by the MTT-method so as to show the effect ofβ-elemene on proliferation of bladder cancer T24 cell lines.The effects ofβ-elemene on cell migration and invasion were detected by Wound Healing assay and Transwell assay.The expression of MMP-9 were detected by Western blot.Results:Bladder Cance T24 cell lines were exposed to theβ-elemene with 0.01,0.02,0.03mg/L for 24h respectively had less absorbance value than control.The proliferation of Bladder Cance T24 cells were inhibited in a dose-depending manner.After being treated withβ-elemene in 0.01,0.02,0.03mg/L for 24 h respectively , and 10 groups were selected randomly.Transwell experiments showed that,the number of control group cells which throughed the Matrigel was 39.2±3.1 and the treated group were 27±1.87,20.2±0.83,14±2 respectively.It means that dependence of dose existed.Using the scratch experiment to observe the migration ability of the Bladder Cance T24 cell lines which were treated withβ-elemene.Observing the healing results on 0,24h of Bladder Cance T24 cell lines exposed to 0.01,0.02,0.03mg/Lβ-elemene,respectively.The results showed that the rate of the control group was significcantly higher than one byβ-elemene,indicating cell migration was inhibited in a dose-dependent manner.Western blot showed that Bladder Cance T24 cell lines were exposed to 0.01,0.02,0.03mg/Lβ-elemene,the expression lever of MMP-9 protein was decreased in a dose-dependent manner.Conclusion:1:β-elemene can inhibit the proliferation,migration and invasion of the Bladder Cance T24 cell lines.2:It is probably one of its molecular mechanisms to inhibit the expression of MMP-9 to inhibit the proliferation,migration and invasion of the Bladder Cance T24 cell lines.
