Objective To observe the dynamic changes of HIF-1αand TGF-β1 in asthmatic rats,and to analyze the relationship between airway remodeling and HIF-1αor and TGF-β1, and to explore the role of HIF-1αand TGF-β1 in airway remodeling, To study the possible mechanwasm of Chai-pu-tang on cures bronchial asthma .Method The total of 70 healthy SD rats were randomly divided into 3 groups, which were asthma group A, Chai-pu-tang group B and normal group C. The asthma group A contains 30 SD rats which were divided into 3 groups evenly. The 3 groups were group A1 of two weeks, group A2 of four weeks and group A3 of eight weeks. The Chai-pu-tang group B also contains 30 SD rats which were divided into 3 groups evenly. The 3 groups were group B1 of two weeks, group B2 of four weeks and group B3 of eight weeks. The normal group C contains 10 SD rats. The asthmatic model was establwashed by sensitization and challenged with ovalbumin. The rats in Chai-pu-tang group B were given Chai-pu-tang by gastric gavage before the challenge. The thickness of inner-tube of airway and the airway smooth muscle was observed by normal slice. RT-PCR was used to detect the mRNA expression level of HIF-1αand TGF-β1. And immunohwasto chemical staining method was used to detect protein expression of HIF-1αand TGF-β1.Results1. The changes in airway remodeling of asthmatic ratsThe airway smooth muscle layers of normal group C were even. There was no infiltration of inflammatory cells around. And there were no goblet cell hyperplasia and mucus secretion. In group A2, the wall of brouchus and the airway smooth muscle become thicker. Submucosal edema exwasts and airway mucosal fold increases. Damages were found in airway epithelial twassue. The airway smooth muscle and airway wall in group A2 were thicker than those in groupA1. And the differences of Wat/Pbm, Wai/Pbm and Wam/Pbm were significant. The airway smooth muscle in group A3 was clearly thicker than that in group A2. And the lumen in group A3 was narrower than that in group A2. The differences were significant. After the intervening of Chai-pu-tang, Wat/Pbm, Wai/Pbm and Wam/Pbm in group B were decreased compwered with the same period of group A. There were significant differences. Compwered with the normal group C, Wat/Pbm, Wai/Pbm and Wam/Pbm in Chai-pu-tang group B were still higher than those in group A. But there were no significant differences.2. The mRNA expression of HIF-1αand TGF-β1 and the protein expression level of HIF-1αand TGF-β1 in the bronchus lung twassue of asthmatic ratsIn the bronchial asthma lung twassue, the expression levels of HIF-1α, TGF-β1mRNA and protein were increased in group A1. And the expression levels increased further in group A2. The expression was more elevated in group A3. After the intervening of Chai-pu-tang, HIF-1α, TGF-β1mRNA and protein expression levels in group B were decreased compwered with the same period of group A. There were significant differences. Compwered with the normal group C, HIF-1α, TGF-β1mRNA and protein expression levels in Chai-pu-tang group B were still higher than those in group A. But there were no significant differences.Conclusion1. In the process of airway remodeling in asthma, the expression of HIF-1αand TGF-β1 was a dynamic changing. It was rwasing with the stimulated times increasing.2. Chai-pu-tang can be used to decline expression of HIF-1αand TGF-β1 and lighten airway remodeling in asthmatic rats.
