ObjectiveThe aim of this study was to investigate the effects of clozapine and olanzapine on body weight, food intake and GLUT4 gene expression.Method32 8-week-old male SD rats were divided into 6 groups:6 in 32mg/kg.d clozapine group,6 in 0.5mg/kg.d olanzapine group,5 in 40mg/kg.d clozapine group,5 in lmg/kg.d olanzapine group,6 in 2mg/kg.d olanzapine group and 4 in control group. All the rats received drugs or water via gavage on 8 each morning. We kept a diary of food consumption each day and recorded body weight every 3 days. After 42 days of treatment, quadriceps femoris of each rat were taken and the levels of GLUT4 protein were detected by Western blot analysis.Result1 the effect on body weight:Control and each experimental groups were matched for body weight before experiment. Neither olanzapine nor clozapine did increase body weight. In contrast, clozapine and high dose olanzapine induced a significant body weight loss. The other experimental groups displayed a non-significant trend. of weight loss.2.the effect on food intake:There were no significant differences in food intake among all the groups at the onset and the end of the experiment. All food intake showed no significant differences.3.The effect on GLUT gene expression:GLUT4 protein expression increased significantly in low dose clozapine group to control group. GLUT4 protein expression in each olanzapine groups and high dose clozapine group showed a non-significant increase compare to control group. ConclusionGLUT4 protein may be involved in the course of clozapine or olanzapine, and the gene expression was increased.
