Objiective: To observe the protection effect of series derivatives of propofol on global cerebral ischemia-reperfusion injury in rats.Method: 78 rats randomly divided into 13 groups, every group have 6 rats, includes nomal, sham, NS,propofol group, CQMU-031 low dose group, CQMU-031 medium dose group, CQMU-031 high dose group, CQMU-032 low dose group, CQMU-032 medium dose group, CQMU-032 high dose group, CQMU-039 low dose group, CQMU-039 medium dose group, CQMU-039 high dose group. The global cerebral ischemia-reperfusion model was established by bilateral common carotid arteries occlusion 20 minutes and haemospasia hypotension in rats, intraperitoneally injection the series derivatives of propofol (equal to propofol 15,30 ,100 mg·kg-1) and propofol (100 mg·kg-1) at the later 30 minutes. Get the neurological behavioral scores by Morris maze test at the 8 days after the surgery . Through the HE staining was determined to see the morphologic changes of neurons in hippocampus.Results: The neurological behavioral scores shows that sham group rats'latency to find platform time isn't significant differnce with normal. But is significant differnce with the CQMU-031-low, CQMU-031-med, CQMU-031-high, CQMU-032-low, CQMU-032-med, CQMU-032-high, CQMU-039-low and NS groups. CQMU-039-med group rats get shorter, CQMU-039-high and propofol group is not significant with normal group. Control to the normal rats'pathomorphological changes of the brain tissue, there is no significant differnce with sham,propofol and CQMU-039-high group rats';but there is significant change in the other group.Conclusion: The I/R model has great hurt on the neurons , delay the latency to find platform time. The derivatives of propofol CQMU-031 and CQMU-032 has no exact nuroprotection on I/R model while CQMU-039 has nuroprotection in rats.PART 2: Protection of Propofol Derivative CQMU-039 on Global Cerebral Iischemia/Reperfusion of RatsObjective: To explore the protect action possible mechanism. of propofol derivative CQMU-039 on rats cerebral ischemia and reperfusion .Method: Through observe the effect of CQMU-039 to global cerebral ischemia/reperfusion on the expression of SOD,MDA,NGF-β,Caspase-3 on hippocampal of rats.Results: Compared with sham group, the propofol derivative CQMU-039'middle and high dose group rats'content of SOD and the protein of NGF-βin hippocampal of rats increased. But the content of MDA and the protein of caspase-3 in the hippocampal of rats decreased.Conclusion: Propofol derivatives CQMU-039 may through improve the body's free radical scavenging activity, inhibition of apoptosis, enhancement of neurotrophic and other areas to reduce the model of global cerebral ischemia-reperfusion brain injury in rats to achieve the effect of brain protection.
