| Objective To investigate the role of heat shock protion 70(HSP70) in the pathogenesis of Graves disease(GD).Methods A total of 25 graves disease patients who admitted in our hospital from March 2010 to July 2010 and a control group of 18 healthy volunteers in the same period were included in this study. Their blood samples were collected.Serum concentrations of HSP70,TNF-a and TRAb in 25 were measured by ELISA , Serum concentrations of malondialdehyde(MDA) were measured by colourimetry , serum concentrations of FT3,FT4,TSH,TPO-Ab,Tg-Ab were measured by hemiluminometry; meanwhile the amount of TLR4 and NF-KB mRNA in peripheral blood mononuclear cell were determined by RT-PCR.Results 1.Serum HSP70,TNF-a and MDA were increased in GD patients [(167.66±100.56 pg/ml vs 42.03±18.46 pg/ml,p<0.05);(27.52±12.09pg/ml vs 7.13±2.62pg/ml,p<0.05);(7.22±0.91 vs 4.04±0.57,p<0.05)].2.Serum HSP70 levels showes correlations with serum TNF-a(R=0.483,P<0.05) and MDA(R=0.412,P<0.05). 3.TLR4 and NF-KB mRNA in peripheral blood mononuclear cell were also higher in patients than in healthy subjects[(0.185±0.006,P<0.05) vs (0.081±0.039,P<0.05);(0.73±0.049,P<0.05) vs (0.31±0.039,P<0.05) ].Conclusion Hsp70 take part in the GD pathogenesis and may play some important roles in the immunopathogenesis of GD. |
