Effect Of Herbal Preparation Kaikoujian On Immune Function Of Mice

Chinese herbal drugs are rarity of traditional medicine, which effective constituents are polysaccharide, glycosides, alkaloid, flavone, mineral substance, etal.Lots of studies confirmed that many Chinese and its components have immune function. However, most of immunopotentiators commonly used in our life ar-e chemical adjuvant, according to that situation, people pay attention to the health, nutrition and food safety,as immunopotentiator,Chinese herbal medicine pos-sess some merits such as safety,low toxicity,efficient effects,no side-effects and non-harmful residue,of which immunity regulate function is the focus of Chin-ese herbal research. In this study, immunity regulates function on mice of Chinese herbal herbal formula Kaikoujian was analyzed. The details are divided into four parts as follows:I To investigate the effect of"Kaikoujian"on the phagocytic activity36 male ICR mice were randomly divided into 4 groups. The animals wer-e orally administered Kaikoujian solution at a dose of 0, 0.0625, 0.125 or 0.25 g /day for 10 days. After that, bodyweight, splenic index, phagocytosis by macrophages were measured. Results indicated that mice administered the herbal solution had increased bodyweight with significant difference for groups of 0.125 and 0.25 g/day herbs (P <0.05) compared to the control, numerically increased splenic indexes (P> 0.05), significantly increased phagocytic activity, clearance index of carbon (P<0.01).II To investigate the effect of herbal preparation"Kaikoujian"on the production of antibodies in mice36 male ICR mice were randomly divided into 4 groups. The animals wer-e orally administered Kaikoujian solution at a dose of 0, 0.0625, 0.125 or 0.25 g/day for 10 days. After that, serum antibody were measured. Reasults indicated that, compared to the control, mice administered the herbal solution had si-gnificantly increased hemolysin antibody levels (P<0.01).III To evaluate the effects of oral administration of "Kaikoujian" on the functions of lymphocytes in mice36 ICR mice were randomly divided into 4 groups. After the animals were orally administered saline solution or"Kaikoujian"solution at lower, medium or high dose for 10 days, samples were collected for measuring lymphocyte stimulation index (SI) by colorimetric MTT assay, expression of IL-4, IL-10, IL-12 and INF-γmRNA by RT-PCR. Results indicated that oral administration of "Kaikoujian" in mice significantly increased splenic lymphocyte SI, expression of IL-4, IL-10, IL-12 and INF-γmRNA.IV To evaluate the effects of oral administration of "Kaikoujian" on the nutritional growth factor levels in mice.36 ICR mice were randomly divided into 4 groups. After the animals were orally administered saline solution or"Kaikoujian"solution at lower, medium or high dose for 10 days, samples were collected for measuring serum insulin -like growth factor-1 (IGF-1) as well as growth hormone releasing peptide (Ghrelin) level by an ELISA kit. Results indicated that oral administration of "Kaikoujian" in mice significantly increased serum IGF-1 and Ghrelin levels.These experiments indicated that herbal formula Kaikoujian has many phar-macological effects, enable to activate immune function of mice. Oral administ-ration of Kaikoujian in mice enhanced the immunity by activating phagocytosis by phagocytic cells, increased cellular immune function and nutritional growth factor levels, enhanced the immunity by activating antibody production.
