Abstract/Summary:
Objectives: 1) To screen del phenotype and determine its RHCE phenotype. 2) To determinate D antigen epitopes for DEL phenotype.3) To determinate D antigen intensity and the positive rate of relative D antigenfor DEL phenotype.Methods:1) To screen RhD negative outpatient and inpatient from hospital in February 2009~June 2010 that screening negative by saline method. 2) To screen Del individual from D negative outpatient and inpatient by IAT and ether absorption elution test. 3) To determine the RHCE phenotype for Del individual by conventional serological methods. 4) The epitope of Red blood cell samples from 74 DEL individuals were analyzed with 16 anti-D monoclonal antibodies specific for 1~9 epitopes of D antigen through minim absorption-elution test to observe the pattern of antigen expression in the erythrocyte membrane, and then to achieve the antigen epitope mapping. 5) To analyse the positive rate of relative D antigen and the intensity of D antigen for the red blood cells of Del individuals by flow cytometry.Results:1)There were 336 RhD negative individuals that screened by saline method from outpatient and inpatient in February 2009~June 2010 . 2) There were 74 DEL phenotype individuals were screened by IAT and ether absorption elution test, occuoy 22%. 3) The RHCE distribution of 74 Del individuals were as follows: Ccee 62 cases accounted for 83.79%;CCee 9 cases accounted for 12.16%;CcEe 2 cases accounted for 2.70%;CcEE 0 cases accounted for 0%;ccEe 1 cases accounted for 1.35%;ccee 0 cases accounted for 0%;ccEE 0 cases accounted for 0%,of which RhC positive phenotype 73 cases accounted for 98.65%. 4) The epitope of Red blood cell samples from 74 DEL individuals were analyzed with 16 anti-D monoclonal antibodies specific for 1~9 epitopes (epD1~6 and epD8~9,7epD was deletion)of D antigen through minim absorption-elution test were all positive. 5) The positive rate of relative D antigen for D(-), DEL, and D(+) were 0.97%±0.18%, 3.06%±0.31%, 99.65%±0.71%, respectively. The intensity of D antigen for the red blood cells of the threre phenotype were 13.89±2.45, 24.18±3.38, 223.32±13.05, respectively.Conclusion: 1) The red blood cells of Del individuals express grossly complete epitopes. 2) There was a significant discrepancy of D intensity between DEL, D(+) and D(-). The D intensity and the positive rate of relative D antigen of Del are slighter larger than D(-) and much less than D(+). Del phenotype individuals have very low antigen intensity and only contain micro number of D antigen molecules.
