Diagnosing ITP By Measuring The CD61 And PAlg With FCM

Objective: The study was aimed to measure three group pepole's(including healthy people group,ITP group and non-ITP group) CD61 and PAIg on the surface of platelet. To evaluate the value of measuring CD61 on the surface of platelet and the platelet-associated antibody PAIg by FCM in the diagnosis and antidiastole of ITP.Method: With FCM,we measured the positive pecentage of CD61, and the positive pecentage and epipolic strength of PAIg(including PAIgA,PAIgM,PAIgG). All people were divided into three groups:healthy people group(30 patients),ITP group(59 patients) and non-ITP group(24 patients).We compared the healthy people group with the ITP and the non-ITP,and did the correlation analysis of PAIg, CD61 and number of platelet in ITP patients.Results: The positive pecentage of PAIgA,PAIgM,PAIgG in ITP group and non-ITP group are (10.93±6.45,16.27±9.89,22.23±15.83)and(13.47±10.32,20.23±9.76,19.82±12.27). The positive epipolic strength of PAIgA,PAIgM,PAIgG in ITP group and non-ITP group are (0.37±0.16,0.45±0.27,0.64±0.51)and(0.42±0.26,0.56±0.32,0.54±0.31). The positive pecentage and epipolic strength of PAIgA,PAIgM,PAIgG in healthy people group are (1.33±0.41,5.04±1.23,5.46±1.43) and(0.23±0.09,0.28±0.11,0.25±0.10).Compare the healthy people group with the other two groups respectively, the positive pecentage and epipolic strength of the two patient groups are both higher(P<0.01),however,the difference is not significant in statistics respectively(P>0.05). The positive pecentage of CD61 in ITP group and non-ITP group and healthy people group are (90.24±13.610),(97.78±1.30)and(99.03±0.67). The positive pecentage of CD61 in the ITP group is lower than the non-ITP group(P<0.05), and there is no significant difference between the non-ITP group and the healthy people group.The number of platelet is negative correlation with PAIgA, PAIgM and PAIgG, and the correlation with PAIgA is the strongest.CD61 lower in ITP group, however, there is no correlation with the number of platelet. The positive pecentage of PAIgA,PAIgM,PAIgG and CD61 in serious ITP group (whose PLT<20×109/L)are(16.27±7.8,24.24±14.57,32.46±16.29) and(82.52±12.82). The positive pecentage of PAIgA,PAIgM,PAIgG and CD61 in mild ITP group (whose PLT>20×109/L)are(8.88±4.47,13.23±5.09,17.85±9.66) and(93.12±12.89).The positive pecentage of PAIg of patients,whose ITP are at high stage,is higher than the patients whose ITP are at low stage.However,the positive pecentage of CD61 is higher in the patients whose ITP are at low stage, and the difference is significant in statistics(P<0.05).Conclusions: The positive pecentage and epipolic strength of PAIgA,PAIgM,PAIgG in both ITP group and non-ITP group are higher than healthy people group . The positive pecentage of CD61 in the ITP group is lower than the non-ITP group and healthy people group. The number of platelet is negative correlation with PAIgA, PAIgM and PAIgG .The positive pecentage of PAIg of patients,whose ITP are at high stage,is higher than the patients whose ITP are at low stage.However,the positive pecentage of CD61 is higher in the patients whose ITP are at low stage, and the difference is significant in statistics.It is valuable to measure the CD61 and PAIg with FCM in the diagnosis, antidiastole and monitoring of ITP.Also,FCM is more sensitive, fast and convenient.
