Objective:To investigate the relationship between HPA-1-6,15 systems genetic Polymorphism and idiopathic thrombocytopenic purpura. Methods:Polymerase chain reaction-specific sequence primers (PCR-SSP) was used to genotype HPA-1-6 and 15 in the ITP group (n=55) and the health control group (n=55);Autoantibodies(GPⅡb/Ⅲa and GPIb/IX) against platelet glycoprotein was measured by a mon-oelonal antibody immobilization of platelet antigen assay(MAIPA) in 55 ITP group and 30 health control group.Finally analyse the HPA-1-6,15 systems genetic polymorphism between auto-antibodies positive ITP patients. Results:1)A11 of the genotypes and allele frequencies were in Hardy-Weinberg equilibrium.The allele frequencies of HPA-1,3 was significantly different between the ITP group and control group (P<0.01).The frequency of HPA-la,3a in ITP group was lower than that in control group,and the frequency of HPA-lb,3b was higher than that in the control group,the frequencies of other alleles showed no significant difference between the two groups(P>0.05).2)31 patients had mono-specific antibodies to GPⅡb/Ⅲa and 10 patients had mono specific antibodies to GPIb/IX.Another 6 patients had antibodies with both antibodies.None people in the health control group was positive of antibodies.There was significant differene between two groups(P<0.05).And HPA-1,3 systems gene showd high heterozygosity in all of the autoantibodies positive ITP patients.Conclusion:There was a possible association between the HPA-1,3 alleles and ITP.
