Due to trauma,tumor,inflammation,aging and other causes of human hard tissue defects, which is the oral medicine and clinical medicine need to be solved problems. The application of the Guided Bone Regeneration technology has brought hope to solve this problem. The use of the barrier membrane is an important technical means in the GBR and to some extent the performance of the barrier membrane determines the success of the GBR technology. Currently barrier membrane materials used in clinical practice can be divided into polytetrafluoroethylene as the representative of the non-degradable material and previous collagen,polylactic acid as the representative of the degradation of materials. Barrier membrane made of non-degradable material requires not only the second operation to remove the membrane, but also is expensive; the barrier membranes made of collagen biodegradable materials do not need the second operation to remove, but there is immunogenicity uncertainty. In recent years selecting the membrane and the ideal material under the membrane joint used in GBR have become the focus of the study. In the experimental the collagen membrane and the autologous periosteum was applicated in the bone defects. By comparing the two materials in the bone defects, it confirmed that the autologous periosteum can promote bone formation and lay the foundation for the preservation of the periosteal in the future clinical application.Objective To evaluate the effect of autologous periosteum in the GBR technique to enhance bone regeneration in dogs and evaluate their osteogenic potential. So to understand the autologous periosteum how to promote the bone healing and the new bone formation.Methods Six healthy dogs were established. In one side of the canine mandibular three 1.0cm×1.0cm sized bone defects were prepared and filled withβ-TCP particles then covered with autologous periosteum or collagen membrane as the two experimental groups and no covering as control. Two dogs were sacrificed at 4, 8 and 12 weeks after operation respectively. Bilateral mandibulars were taken as specimens.The new bone formation of the experimental group and the control group in diffenent periods of time was observed by means of general observation, environmental scanning electron microscope observation, X-ray examination and histologic analysis as well as the suface change of the bone defects and the surrounding tissue reaction.Results There were varying degrees bone regeneration in all experimental sides 4,8,12 weeks after operations, while only a small amount of bone regeneration in the control group. In the 4 weeks'autologous periosteum group and collagen membrane group, there was a little degradation of the material and new bones formation was not mature.No degradation of the material was obvious and few new bones were formed in the control group in 4 weeks. In the 8 weeks' control group there was soft tissue in defects. In the experimental groups part of the materials were degradated and the formation of new bones formation was more than the control group. In the 12 weeks'autologous periosteum group and collagen membrane group, degradation of the material was almost completed and new bones formation was mature. The material was partly degraded and new bones were formed but not mature in the control group in 12 weeks. Conclusion(1) Autologous periosteum used as barrier membranes in GBR technique have good biocompatibility, no immunogenicity and have the promotion of bone healing and new bone formation. For due to trauma, tumor, inflammation, aging and other causes of human hard tissue defects provide an objective basis and provide experimental evidence to find an alternative membrane material for the GBR technique to repair bone defects around the implants.(2) Tricalcium phosphate as a filling material under the membrane can maintain the existence of tissue regeneration of the space and fixed blood clots.β-tricalcium phosphate combined with autologous periosteum were used in GBR technique .The synergy between the two effectively promote bone repair and acceleration of bone remodeling.
