Solidago canadensis belongs to genus Solidago L. (Asteraceae). It is a native plant of South America and now distributes widely in South China. It has been used as antioxidant, anti-inflammatory, antimicrobial and antitumer agents in Europe for about 700 years. So far, polyacetylene, diterpene, saponins, flavones, phenylpropionic acids were isolated from this plant. But few studies were carried out on S. canadensis collected in China. In our study, phytochemical investigation resulted in the isolation of a new diterpene, together with 13 known compounds. Meanwhile, cytotoxicity activities of four diterpenes were evulated.Object: To study the chemical constituents of S.canadensis.Besides and to primary pharmacological effect on cytotoxicity of all of the diterpenes.Methods: The whole plants of S. canadensis were percolated with 95% EtOH and partitioned into chloroform, ethyl acetate, n-butyl alcohol successively. Constituents were isolated and purified by silica gel column chromatography, Sephadex LH-20 column chromatography, TLC preparation and recrystallization. Their structures were identified on the basis of their chemical and physical properties as well as the spectra data of ESI-MS, 1H-NMR and 13C-NMR et al. Study the cytotoxicity of four diterpenes. SMMC-7721, MCF-7, DU-145 and LH-60 were selected as study objects. MTT was used to determine the effect of these compounds on the cells. Inhibition rate was used to observe cytotoxic activity.Results:14 compounds were isolated from S. canadensis, which were identified as 6-Hydroxy-cleroda-3,13-dien-15,16-olide (Ⅰ), 2-Hydroxy-6-methoxybenzoic acid (Ⅱ), 3-Formylindole (Ⅲ), 3β,4α-Diol -6β-tigloyloxy-cleroda-13Z -en-15,16-olide (Ⅳ), 3β,4α-Diol -6β-angeloxy-cleroda-13Z -en-15,16-olide (Ⅴ), solicanadene A (Ⅵ), Olean-12-ene (Ⅶ), Urs-12-ene (Ⅷ), Stearic acid (Ⅸ),α-Spinasterol (Ⅹ), Kaemferol (Ⅺ), Quercetin (Ⅻ), Kaempferol-3-O-β-D-rutinoside (ⅩⅢ), Rutin (ⅩⅣ)。From the observation of four compounds from S. canadensis on cytotoxic function experimental, there was a dose-effect relationship, but compounds IV, V and VI almost did not have inhibition to SMMC-7721, MCF-7, DU-145 and LH-60. Conclusion: Compound VI is a new compound.CompoundsⅡ,Ⅲ,Ⅳ,Ⅴ,Ⅶ,Ⅷ,Ⅹare isolated from S. canadensis for the first time.CompoundsⅣ,ⅤandⅥdid not have cytotoxic activities at a concentration of 100μg/ml.
