Objective: Through the pre-experimental research work, We observed that experimental animals,the general voltage electric burns specimens mesenteric, intestinal mucosa, kidney cortex, liver, gastric mucosa, deep fascia, the soft meninges and other micro-circulation changes.We can find not only the high-voltage caused by peripheral tissue microcirculation disturbance,but also to distant organs,caused by microcirculation disorders,leading to systemic microcirculation,causing body damage can be observed that the performance of leukocyte rolling and adhesion along the vascular,white small thrombus mobility and erythrocyte aggregation phenomena. It is proved that immunoglobulin super family (lg-SF) adhesion molecules appear increase after high-voltage burns serum.We designed this experiment to observe the continuous changes of ICAM-1,VCAM-1,PECAM-1 on pulmonary tissue in voltage electric burn to further explore the mechanism microcirculation disorder,and intervention of pentoxifylline,These microcirculatory disturbance after burn injury prevention and treatment for high-voltage and provide a theoretical basis for the greatest possible reduction in patient mortality,improve survival rate.Methods:1 Group: 180 healthy grown up male S-D rats are randomly divided into three groups, high-voltage electrical burned experiment group (experiment goup), high-voltage electrical burn PTX reatment group (treatment group)and fake high-voltage electrical burn group (control group), 60 rats of each group. The time phase of these groups are divided into six different phases, ahead(T0), immediately(15min) (T1), 1h(T2), 2h(T3), 4h(T4), 8h(T5), 10 rats of each group.2 Preparetion:to weigh the rats,number them,The experimental drug concentration required by experiment dubbed.3 Duplication of high-voltage electric burn model: Anaesthetic rats of electrical burn experiment group and PTX treatment group are burned by the 10,000 volts high-voltage electric system for 3 seconds,they are duplicated full-thickness electrical burn with body tissues that touch with the electrodes, we immediately inject PTX(50mg/ml) to rats of treatment group through enterocoelia,normal sodium to the experiment group and the control group. The progress of control group is the same as the electric burn group,but there isn't electric current in the body of control group rats.4 Collection and detection:In different time phases open chest to pick Left ventricular organization from rats'heart, made paraffin section, and preserve them in 4℃refrigerator.5 The pulmonary tissue content of ICAM-1,VCAM-1,PECAM-1 of the rats were measure by immunohistochemisty (IHC) at different time phases during early electrical burn.6 Analysis of Statistics: statistically analysis all the data with SPSS13.0.all the dates should be expressed with x±s.we apply two factors factorial design of variance analysis among groups. It has significant statistically meaning if p<0.05.Results: the changes of ICAM-1,VCAM-1,PECAM-1 in the pulmonary tissue1. The pulmonary tissue levels of ICAM-1,VCAM-1,PECAM-1 haven't obviously change within the control group.(p>0.05)2.High voltage electric burn is not the same time, the experimental group of early-phase pulmonary tissue ICAM-1,VCAM-1,PECAM-1 levels and the high voltage electric burn higher than before the ICAM-1,VCAM-1,PECAM-1 levels (p <0.05); in which ICAM-1 expression level increase at 15 minutes after burn from the high-voltage and continues to 8 hours; VCAM-1 expression level increase at 15 minutes after burn from the high-voltage, reachs a peak at 4 hours after burn and continues to 8 hours; PECAM-1 expression 4 hours after injury, reached a peak later with the down to the low level of time-varying。3. High voltage electric burn after PTX treatment group was not accompanied by an early-phase pulmonary tissue ICAM-1,VCAM-1,PECAM-1 and electricity before the higher pulmonary tissue ICAM-1,VCAM-1,PECAM-1 levels (p <0.05), but at the same time compared to electric blow pulmonary tissue ICAM-1,VCAM-1,PECAM-1 levels decreased to varying degrees (P<0.05); in which VCAM-1 levels from 8 hour after injury,and PECAM-1 after 2 hours, since high expression; pulmonary tissue ICAM-1,VCAM-1,PECAM-1 levels in 8-hour-phase electric shock group and treatment groups decreased between the two groups, with significant difference (p< 0.05).Conclusion:1 ICAM-1,VCAM-1,PECAM-1 have increase after high-voltage electrical burned experiment group and PTX treatment group,play an important role in microcirculatory disorder.2 The study indicates ICAM-1,VCAM-1,PECAM-1 of PTX treatment group higher than experiment group, but lower than the control group still. PTX might be helpful in preservation ICAM-1,VCAM-1,PECAM-1 and relief microcirculatory disoeder after high-voltage electrical burn.
