Leaves of Baphicacanthus cusia processed into Qingdai is the course of transformation for precursors in Qingdai into indigo and indirubin,including three links of soaking,making and refining.This paper studyed on the processing principles,processing technology,quality standards of Qingdai systematically,lay the foundation for controlling the processing of Qingdai omnidirectional and making the industrialized production realized.To provide a scientific evaluation indicators for studying the mechanism of Qingdai processing,this paper amended the description,identification,determination of Qingdai respectively. Description:It is dark bule powder,light and puffy or irregular and porous masses,which is finely powdered on twisting.Odour,slightly grassy;taste,weak.Identification:Mobile phase is the mixture of methylbenzene-chloroform-acetone (5:4:1) instead of benzene-chloroform-acetone (5:4:1).Determination:The method is simultaneous determination contents of Indigo and Indirubin in Qingdai by HPLC.The results showed that purified by the foam flotation process,the Qingdai particles and powder is consistent with the requirements of《Chinese Pharmacopoeia》(2005 edition). Instead of benzene as the mobile phase,methylbenzene has less impact on the TLC.Meanwhile,indigo and indirubin spots were clearand and the separation was good.This simultaneous determination is scientific,accurate and convenient. The amendments of description,identification and HPLC method of indirubin determination has included by《Chinese Pharmacopoeia》(2010 edition).This paper using the methods of HPLC-ELSD,pNPG and comparison studyed the traditional immersion processing of the leaves of Baphicacanthus cusia that indican released from the leaves,hydrolyzed byβ-Glucosidases simultaneously.Using Plackett-Burman design and single factor experiment,we screened the significantly influencing factors in soaking and making links,clarifed that the soaking temperature,oxygen content before making indirubin and pH when making indirubin were the main factors generating indirubin.Using Box-Behnken design response surface analysis method optimize the soaking and making indigo,indirubin process of Qingdai separately.The soaking and making indigo process of Qingdai was identified as the fresh leaves soaked in the water,immersion pH 3.5,temperature 25℃,solvent volume-leaf mass (mL:g) 13,soaked in dark,soaking 24h,then filtered to remove leaf residue,filtrate plus ammonia water to adjust pH to 9,ventilated 30 min,plus 10% of fresh milk of lime(taking CaO which mass is 10% of fresh leaves,adding boiling water which the amout is 10 times,stirring constantly to form suspension),stir well and put it aside for 24h,filter to take the precipitation,the precipitation-drying is crude Qingdai.The soaking and making indirubin process of Qingdai was identified as the fresh leaves soaked in the water,immersion pH 7,temperature 60℃,solvent volume-leaf mass (mL:g) 15,soaking 48h,then filtered to remove leaf residue,filtrate ventilated 30 min,plus ammonia water to adjust pH to 10.5,then ventilated 180 min additional,plus 10% of fresh milk of lime(taking CaO which mass is 10% of fresh leaves,adding boiling water which the amout is 10 times,stirring constantly to form suspension),stir well and put it aside for 24h,filter to take the precipitation,the precipitation-drying is crude Qingdai.Through the reaction mechanism,this paper screened important factors,optimized processing technology,studying traditional processing systematicly by three levels of mechanism,factors and process. We regulated the direction of production of indigo and indirubin according to market demand.Meanwhile,we established "M-P-T-I" new model of product development,which provide a model for other traditional Chinese medicine.
