| Objective Aim To investigate the association microRNA-22,microRNA-382, microRNA-134 and microRNA-206 polymorphisms with genetic predispostion and early recurrence after resection for HBV-related hepatocellular carcinoma(HCC).Methods A total of 378 subjects, including 128 cases with HBV-related HCC, 121 patients with chronic hepatitis B and 129 normal controls, were enrolled in this study. We used bioinformatics methods to predict and select out a total of nine tag SNPs in miRNA-22, miRNA-382,miRNA-134 and miRNA-206 gene promoter regulatory region, matrix assisted laser desorption ionization time of flight mass spectrometry analysis was executed genotype the nine SNPs, including rs11078597rs6502892 in miRNA-22, rs1951032 rs4906032 in miRNA-382,rs12886869 rs4143957 in miRNA-134 and rs16882131 rs69020648rs945210 in miRNA-206 gene. And calculated the association betweengenetic variants and the risk of HCC, and assessed the impact of mutations on HCC early recurrence.Results The distributions of genotypes and allele of miRNA-134rs4143957 and miRNA-206 rs16882131 were significantly different between HCC,CHB and CON the three group. However, the CC genotype and C allele of miRNA-206 rs16882131 was associated with reduced risk of HBV-related HCC(OR=0.354,95% CI 0.163-0.771;OR=0.182,95% CI0.124-0.267);the TT genotype and T allele of miRNA-134 rs4143957 was associated with reduced risk of HBV-related HCC in males(OR=0.180,95% CI 0.062-0.527; OR=0.465,95% CI 0.295-0.734). And miRNA-134 rs4143957 was related to the early reccurence of HCC,miRNA-134 CT genotype may be a protective factor for recurrence of hepatocellular carcinoma after operation.Conclusion miRNA-134 rs4143957 and miRNA-206 rs16882131 polymorphisms may be associated with the genetic predispostion of HBV-related HCC. Moreover, miRNA-134 rs4143957 polymorphisms may also be associated with the early reccurence of HCC, and miRNA-134 CT genotype may be a protective factor for recurrence of hepatocellular carcinoma after operation. |
