Objective:1.To investigate the effects of chronic forced swimming stress on the behaviorals change and the expression of GKAP in the brain of rats .2.TO observe the intervention of the non-competitive N-methyl-D-aspartate (NMDA) receptor antagonist MK801 on the reaction to the chronic forced swimming stress.3.TO probe the prossible central nervous mechanisms of chronic stress reaction.Methods1.Thirty SD rats were randomly divided into three groups:Control group ,swimming stressed group and swimming stressed+MK801 group,with ten in each group,the cotrol group was under normal feeding, the swimming stressed group accepted ten minutes forced swimming stress everyday,and the swimming stressed+MK801 group accepted subcutaneous injection of MK801 before stress, the forced swimming last 14 days.2.Weight of all rats was measured on the 1rd,6th,10th and 14th day after the beginning of experiment.The immobile time of each rat in forced swimming stress groups was recorded.Liquid consumption of each rat was assessed in the following 4 days after forced siwmming finished.3.The GKAP level in hippocampus,prefrontal contex and basal ganglia after stress was measured with the Western-blot.Results1.After forced swimming,compared to the controls and swimming stressed+MK801 group ,the weight growth rate, sugar consumption and sugar preference percentage in the swimming stressed group had significantly less numbers(p<0.05),there was no significantly defference between the rest groups.Also,the immobile time of rats in the forced swimming group was significangly more than another group (p<0.05),and it had significantly defference between the 1rd day and the 14th day (p<0.05).2.The level of GKAP was significangly higher in hippocampus of the swimming stressed group (p<0.01) . However there was no significangly defference among three groups in prefrontal contex and basal ganglia.3.The level of GKAP in hippocampus of the swimming stressed group had positive correlation with immbile time(p<0.01),but had negtive correlation with weight growth rate and sugar consumption(p<0.05).Conclusion1.Chronic forced swimming stress can be established as chronic stress depression model successfully.2.GKAP in hippocampus may paticipated in the central nervous mechanisms in the depressed reaction of chronic stress.3.MK801 had potential antideprssant effect.
