| Background and aims: She ethnic population is one of the minorities in China, and there is limited data about the type 2 diabetes mellitus (T2DM) susceptibility genes in this population. Therefore, we conducted this study to detect the relationships of FTO (rs8050136) and SLC30A8 (rs13266634) genes with T2DM.Methods:All subjects in this study were unrelated She population in Ningde city of Fujian province, including 142 subjects with T2DM, 336 subjects with impaired glucose regulation (IGR) and 515 subjects with normal glucose tolerance (NGT). Physical measurements were performed and clinical biochemistry data was collected. Genomic DNA was extracted from peripheral blood leukocytes, and genotyping was performed with ABI 7500 system based on Taqman probe technology. Data analyses were conducted with Windows SPSS 13 statistical software package (SPSS, Chicago, IL, USA).Results: (1)The frequencies of AA genotype and A allele in FTO (rs8050136) were both higher in T2DM and IGR groups than NGT group. Compared C allele carriers, subjects who carrying A allele was found to be 1.679 times (odds ratio[OR]=1.679,P<0.001) more risk for T2DM and 1.382 times (OR=1.382,P=0.001) risk for IGR. (2) The frequency of TT genotype in SLC30A8 (rs13266634) in T2DM group was significantly lower than that in IGR and NGT groups, while the frequency of CC genotype was higher than that in IGR group. Compared with T allele carriers, subjects who carrying C allele was found to be more risk for T2DM (OR=1.472,P=0.006),but the risk for IGR was not significan(tOR=0.898,P=0.283). (3) After adjusted for sex, age and body mass index (BMI) , we found AA genotype carriers of FTO had lower level of Homa-βcompared with CC genotype carriers, and also the Homa-βlevel was lower in CC genotype carriers of SLC30A8 than TT genotype carriers . (4) After adjusted by age, sex and BMI, logistic analyses revealed that rs8050136 of FTO were both significantly associated with T2DM (AA vs. CC:OR=1.707, P<0.001) and IGR (AA vs. CC:OR=1.334, P=0.009), while rs13266634 of SLC30A8 was associated with T2DM (CC vs. TT:OR=1.478,P=0.009) but not significantly associated with IGR (CC vs. TT:OR=0.867, P=0.176). (5) The frequencies of genotypes and alleles in FTO (rs8050136) were not found to be significantly different between obesity/overweight group and normal body weight group. Logistic analyses revealed that there was no significant relationship between FTO variance and obesity/overweight after adjusted by age and sex (AA vs. CC:OR=1.117,P=0.332).Conclusions: Polymorphisms in rs8050136 of FTO and rs13266634 of SLC30A8 were both significantly associated with T2DM in She population,and their variants might influenceβ-cell function. However, FTO gene might not significantly associated with obesity/overweight, in order to confirm this relationship, much lager simple size must be needed. |
