Objective:This study was designed by observing the impact for Aβ-42 and IDE in AD brain tissue by electro-acupuncture therapy. And reveal the role of acupuncture effective in treating AD. Furthermore, it can provide more experimental evidence for acupuncture clinic treatment of Alzheimer's disease (AD).Method:Experimental animals were randomly divided into four groups: A group(sham-operated group),B group(Model group),C group (Western medicine group) and D group (EA group).Except A group, all of the animals need to be modeled by intracerebroventricular injection of STZ(ICV-STZ). The sham-operated group were given double distilled water respectively. Each group a different approach.After curing for 2 months,we use Morris Water Maze system to check the ability of animals about learning memorizing at first.Then we need to kill the animal and take out the specimen. Moreover, we need to use immunohistochemical technique to check the change of Aβ-42 and IDE in brain tissue,then analyze it.Result:After the subjective observation, we found that treated group had better behavior about action and reaction than other groups. The result of Water Maze shows that:the escape latency of model group was longer than other groups,but the swimming time in the third quadrant was less than other groups.Meanwhile,the EA group and the sham group was not significant.Index test results show:In model group,the levels of Aβ-42 were significantly elevated in rat brain (P<0.01), while the levels of IDE were decreased (P<0.01).After treatment,the levels of Aβ-42 was significantly lower (P<0.01),IDE expression was significantly increased in EA group (P<0.01).And western medicine group was no significant difference (P>0.05).Conclusion:EA can improve the ability of rats about learning memorizing,and prevent the Alzheimer's disease (AD).On the other hand, it can reduce the levels of Aβ-42 in brain tissue and improve the activity of IDE.Furthermore,it can reduce the generation and deposition of Aβ-42,it also can reduce the neurotoxicity of Aβ-42.Then it will inhibit the formation of senile plaques and improve symptoms related to AD.
