Oncomelania hupensis Gredler is the only intermediate host of Schistosoma japonicum Katsurada, so to eliminate Oncomelania hupensis Gredler by molluscacide is an effective method to control the prevalence of schistosomiasis..Niclosamide(Nic) is the only molluscacide which was recommend by WHO and frequently-used in China. But there are some economy and echnology drawbacks such as low solubility in water weak selective toxicity and expensive to the government. So newtype molluscacide shoud be researched according to governmental important technique needs. This thesis use NiC and water-soluble steroidal saponins/alkaloids in water extraction of plant as test drug, and use Oncomelania hupensis Gredler and Ophicephalus argus as target organism, to investigate the effect of drug to the enzymatic activity and protein expression of target organism. The result can provide a basis for the screen of high activity lead compound and particular target protein.1,The study of optimal catalytic conditions of glutamic-oxalacetic transaminase (GOT)from the soft body of Oncomelania hupensis and liver of Ophicephalus argus.The optimal catalytic conditions of GOT was established by L25(56) orthogonal matrix method and range analysis.When enzyme concentration was 0.2/0.4 mg/ml, substrate concentration was 5.0/5.0umol/L, pH was 7.5/8.0, reaction time was 60/20min and temperature was 45/45℃, respectively,the amino transamination rate was soonest. The result show that GOT from different species have different catalytic activity, This can provide the basis for the effection to GOT activity by test drug.2,The effects to GOT activity by incubating with drug/being exposed in drug solution.2.1 The relationship between GOT activity from Oncomelania hupensis and test drug concentration by incubatingThe concentration effect:GOT was incubated with NiC and extraction from Reineckea carnea Kunth,Solanum xanthocarpum schrad. et Wendl,Dioscorea zingiberensis C. H. Wright at different concentrations for 20min,when concentration was 0.25,0.5,0.5 and 2.0mg/L.The induction rate was highest up to 19.3,23.7,18.9 and 11.52%respectively.The result show that GOT was most sensitive to steroidal alkaloids extracted from Reineckea carnea.2.2 The influence on GOT activities from Ophicephalus argus liver by incubating with NiC. The concentration effect:GOT was incubated with NiC at different concentrations for 20min,when drug concentration was 0.125mg/L,the induction rate was highest up to 39.94%.The effect of time:GOT was incubated with 0.25mg/L NiC for different time,when incubated for 40min, he induction rate was highest up to 44.86%.2.3 The influence on GOT activities from Ophicephalus argus liver by being exposed to different concentrations of NiC.Ophicephalus argus was exposed to different concentrations of NiC, the organism was only living for 3.5 hours in 1.5mg/L NiC solution,and the induction rate of GOT activity was highest up to 113.99%.This results show that GOT catalytic activity induced by NiC maybe one of the causes leading to death.3,The effect to protein expression of Ophicephalus argus by being exposed to NiCTake the liver after exposed to 0.5mg/L NiC for 48h. Total liver proteins were extracted and separated by 2-DE and analyzed by PDQuest 8.0.1,10 proteins expression were significantly up regulated. The result can provide a basis for the toxicity mechanism and potential target protein of new molluscicides.
