Schistosomiasis is a widely-spread parasitic diseases on the earth only second to Malaria.As Oncomelania hupensis is the only middle host of Schistosoma,the elimination of Oncomelania hupensis can effectively put down the spreading of Schistosomiasis.Nic is the popular chemical anti-Oncomelania hupensis drug,but is slow in action,pricy and is highly poisonous to the fishes.Plant molluscicide drug has received attention because of such technical and budget-related advantages as being lowly poisonous,easy to biologically deteriorate,and highly affordable.In this thesis,I study the quantity of activity ingredients and mechanism of action of a new plant molluscicide product,and the main results are as follow:1.The establishment of quantitative analysis of the active ingredients SX-Ⅰof product with high-performance liquid chromatography(HPLC).Methods The mobile phase was the mixture of Methanol-Acetonitrile-Ammonium acetate(V/V/V,30:60:10). C18 column was used.Flow rate was 1.0 mL/min and the wavelength of UV detection was at 210 nm.Results The linear range of SX- I was from 100 to 500μg/mL and the average rate of return was 108.8%,RSD was 0.14%.Conclusion The method is simple and accurate which can be used for the quality control of the active ingredients SX-Ⅰ.2.Study the induction of NM and SX-Ⅰplant molluscicide drug on Oncomelania hupensis enzyme and protein.Methods HPLC test,PAGE and SDS-PAGE electrophoresis test.Results The activity of Oncomelania hupensis AchE and GOT is increase first,then steadily decrease,while being immersed by NM and SX-Ⅰ. Conclusion The solution of NM and SX- I can kill Oncomelania hupensis by breaking the balance of regular particle metabolism and blocking the synthesis of protein,thus eventually resulting in the death of Oncomelania hupensis.3.Tested the effect on Oncomelania hupensis AchE of SX fruit powder,AC seed powder and CuSO4 mixed with different concentration.Methods Testing the sensitivity of AchE with regard to drugs using improved Ellman method.The statistical methods like L25(53) orthogonal analysis and range analysis have been employed.Three factors and five levels are determined by experiments.Combinations with the highest enzyme activity inhibition rate are tested.Results The optimal combination of molluscicides mixed on Oncomelania hupensis AchE activeness is:25 mg/L SX fruit powder,16 mg/L AC seed powder and 2 mg/L CuSO4.Conclusion This method is easy and effective,can serve as a reference for developing new Oncomelania agents which are highly poisonous for Oncomelania hupensis and safe for fishes.
