| The drug couple is the most foundational and simplest form in the prescription of traditional Chinese medicine. It only contains two relative fixed Chinese medicines. Although it simplicity, the drug couple implies the central principle of composed with traditional Chinese medicine:the "seven niv" theory and it form the base of the complex prescriptions. The drug couple is effective, easy to use and have been testified for generations. Because it has the characteristics of complex prescription, but simple enough for modern research. The drug couple provides a unique viewpoint to investigate the prescription compatibility in traditional Chinese medicine.Cortex Phellodendri and Rhizoma Anemarrhenae is one of clearing virtual hot medicine which is very commonly used in clinical. The earliest appearance of this couple is in the Zishen Pellet fromwritten by Li GAO in Jing-dynasty. Now the pharmacological research shows, berberine in cortex phellodendri's total alkaloid is the main ingredient which can improve renal dysfunction in diabetic nephropathy rats. Rhizoma Anemarrhenae saponins can greatly improve the glucose tolerance of experimental rats; and reduced fasting hyperglycemia in experimental diabetic mice. We choose the Rhizoma Anemarrhenae and Cortex Phellodendri drug couple as the object to investigate, in order to reveal its application principle, and provide method and basis for further development of research both on modern medicine preparation and material foundation for effective parts.In this paper reviewed the present research on the couple of Cortex Phellodendri and Rhizoma Anemarrhenaem comprehensively. On this solid base, this paper study the content determination of Cortex Phellodendri and Rhizoma Anemarrhenae's altogether extraction; researching the extraction and purification procedure; the quality standard of altogether effective parts extract from Cortex Phellodendri and Rhizoma Anemarrhenae mixture; and investigating its capability of lowering blood sugar and lipid. 1. Investigating the content determination of the drug couple's altogether extraction:Using berberine hydrochloride, phellodendrine, timosaponin BII for reference, determine the amount of total alkaloid, total saponins by UV spectrophotometry and HPLC to quantitate the berberine hydrochloride, phellodendrine, timosaponin BII in altogether extraction 2. The extraction and purification process of altogether effective parts:total saponins& total alkaloid.This study goes further based on GuanGongHui's research. During the study, using single-factor test method to inspects different factors such as extraction time and solvent amount, as well as the ethanol concentration. Finally established the best extraction procedure. Purification process research using macroporous resin to enriching and purify the total saponins & total alkaloid. Screening different types of resin inspected their static and dynamic adsorption performance, and the concentration and flow of sample solution, elution solvent concentration. After trials the best purification process is established:Use AB-8 resin to purified total saponins & total alkaloid; raw material and dry resin weight ratio is 1:1, sample liquid concentration on crude drug for 0.05 g/mL, with 10BV distilled water, in turn 10BV70% ethanol with 1～3mL/min, collect the fraction of 70% ethanol elution part, vacuum drying, get the purity of the effective group is 50.0% or above. 3. Quality standard of altogether effective parts:Using berberine hydrochloride, phellodendrine, timosaponin BII as reference to establish a TLC identification method for altogether effective parts. Create a UV quantitative method to determine the content of total saponins& total alkaloid in altogether effective parts. Use HPLC to quantitative the berberine hydrochloride, phellodendrine, timosaponin BII in altogether extraction. Preliminary established quality standard for altogether effective parts. 4. Altogether effective parts lowering blood sugar & lipid research:Use STZ to create hyperglycemia modeling on mice. After some course of treatment, the altogether effective parts extract from Cortex Phellodendri and Rhizoma Anemarrhenae does have some effect on dropping blood glucoseUsing high fat feed to cause lipid metabolism disorders on obesity rats. After some time the altogether effective parts does not show any distinct effect on cholesterol-lowering. |
