Objects1. To evaluate the clinical effect and security of Fufang Tufuling granules on Hyperuricemic patients, by observing the changes of indexes, including uric acid, clinical symptoms and indicators of safety.2. To investigate the effects of Fufang Tufuling granules on uric acid level in Model of Hyperuricemic Mice that using adenine and Oxonic Aeid to prove its effectiveness.3. To investigate the effects of Fufang Tufuling granules on mouse renal function through contrasting the level of creatinine and urea nitrogen.4. To investigate the effects of Fufang Tufuling granules on mouse uratic renal injury by analyzing the change of kidney.5. To approach the protecting mechanism of Fufang Tufuling granules on mouse renal function by analyzing the level of IL-1β,IL-6 of renal homogenate.Methods1.60patients of Hyperuricemia were randomly divided into Fufang Tufuling granules group, Benzbromarone group. The patients in Fufang Tufuling granules group were treated by concentrated granules of Fufang Tufuling granules. The patients in Benzbromarone group were treated by Benzbromarone tablets. There were 20 days for a course of treatment. The clinical symptoms, physical signs and laboratory data before and after treatment were noted and compared.2.60 Kunming male mice of SPF grade were randomly divided into High-dose Fufang Tufuling granules group, Medium-dose Tufuling granules group, Low-dose Tufuling granules group, Model group and Control group. All mice were established with adenine method combined with uricase inhibition method to build hyperuricemic nephropathy model except Control group, and the corresponding drugs were administrated at the 7th day. On the 22nd day, the eyeballs of mice were removed and the blood was obtained to detect uric acid creatinine and urea nitrogen levels. Simultaneously, the kidneys were removed to detect the level of IL-1β,IL-6 by Elisa and observe structure change of the mouse.Results1. The significant decrease of the degree of uric acid was found in the two groups of Hyperuricemic patients after treatment (P<0.05). The total effective rate was 86.21% in Tufuling granules,92.86% in Benzbromarone group, There was no statistically significant difference between Tufuling granules and Benzbromarone group. The effect in these two groups was similar.2. Imposed by different factors to each group, oral administration of Tufuling granules could significantly reduce the uric acid in mice in a dose-dependent manner, and it was significant difference than that in Model group (P<0.05). The effects of High-and Medium-dose Tufuling granules group were similar with Benzbromarone group (P>0.05), while the effect of Low-dose Tufuling granules group was weaker than Benzbromarone (P<0.05).3. Imposed by different factors to each group, oral administration of Tufuling granules could significantly reduce the creatinine and urea nitrogen in mice in a dose-dependent manner, and it was significant difference than that in Model group (P<0.05). The effects of High dose Tufuling granules were similar with Benzbromarone group (P>0.05), while the effect of Medium and Low-dose Tufuling granules was weaker than Benzbromarone (P<0.05).4. During the experiment the Control group's body weight keep on a rise, the Model group's body weight keep on decrease, after experiment, the Tufuling granules and Benzbromarone group's body weight both have a rise, and it was significant difference than that in Model group (P<0.05). The effects of High-and Medium-dose Tufuling granules group were similar with Benzbromarone group (P>0.05). 6. Tufuling granules can repair the pathological impairment if the kidney, under observation of photoscpe and electeic scope, the effection of High-dose is the best. Results1. Tufuling granules could reduce the uric acid and improve a part of clinical symptoms effectively in Hyperuricemic patients, while it is safety.2. Tufuling granules can rise the weigth of model mouse.3. Tufuling granules can improve the model mouse's renal function.The mechanism of reducing uric acid might be the action of inhibition to the IL-1β,IL-6 expressions.
