Comparison Of Role Of Several Medicines In Suppressing Bleomycin-induced Pulmonary Fibrosis In Rats

Objective:To investigate the role of dexamethasone,NAC,IFN-y and LMWH in suppressing bleomycin-induced pulmonary fibrosis in rats.Method:One hundred and fifty SD rats were randomly divided into six groups(25 rats in each group), ie. a normal group,a bleomycin-induced pulmonary fibrosis model group,a dexamethasone-treated group,a N-acetylcysteine-treated group,a Interferon-gamma-treated group,a low molecular weight heparin-treated group.Five rats in each group were randomly killed on lth day,3th day,7th day,14th day and 28th day,respectively,and lung tissue samples were harvested for histopathology study. HE and Masson staining were used to determine the extent of alveolus inflammation and pulmonary fibrosis,respectively. Histoimmunochemical stain was used to determine theprotein levels of transforming growth factor-(31 (TGF-β1), Serum and bronchoalveolar lavage fluid(BALF) were harvested for determine concentration of TGF-(31 by means of ELISA, lung tissue samples were harvested for determine concentration of hydroxyproline(HYP).Results:1 Stopathological study showed that treatment with dexamethasone-treated group,NAC-treated group,INF-γ-treated group and LMWH-treated group remarkably meliorated the extent of alveolus inflammation and suppressed pulmonary fibrosis on 28th day(compared with model group, P< 0.05). Stopathological study showed that extent of alveolus inflammation and pulmonary fibrosis was not significant difference between the four drug intervention group on 28th day2 Histoimmunochemical study suggested that dexamethasone-treated group, NAC-treated group,INF-y-treated group and LMWH-treated group could inhibit the expression of TGF-β1 on 14th day and 28th day (compared with model group,P<0.05),and the expression of TGF-β1 was more obvious in NAC-treated group and LMWH-treated group than those in dexamethasone-treated group on 28th day (P<0.05); Histoimmunochemical study suggested that expression of TGF-β1 was not significantly different between NAC-treated group,INF-y-treated group,LMWH-treated group(P>0.05).3 ELISA study suggested that dexamethasone-treated group,NAC-treated group INF-γ-treated group and LMWH-treated group could inhibit the expression of TGF-β1 in serum on 7th day,14th day and 28th day(compared with model group,P<0.05),and bronchoalveolar lavage fluid(BALF) as that in the tissue, and the suppression of TGF-β1 was more obvious in NAC-treated group on 14th day and 28th day and LMWH-treated group on 14th day(compared with dexamethasone-treated group group,P<0.05); ELISA study suggested that expression of TGF-β1 in serum and BALF was not significantly different between NAC-treated group,INF-γ-treated group,LMWH-treated group(P>0.05).4 The treatment with dexamethasone,NAC, INF-y and LMWH remarkably reduced contents of hydroxyproline(HYP) of lung tissue on 7th day,14th day and 28th day (compared with model group, P<0.05), and the suppression of contents of HYP was more obvious in NAC-treated group(on 14th day and 28th day) and LMWH-treated group(on 28th day) than those in dexamethasone-treated group (P<0.05); The suppression of contents of HYP was not significantly different between NAC-treated group,INF-y-treated group.LMWH-treated group(P>0.05).Conclusions:1 The expression of TGF-β1 is a small amount in the normal lung tissues,and the expression of TGF-β1 is significantly increased in bleomycin-induced pulmonary fibrosis model. 2 Dexamethasone,NAC,IFN-γand LMWH can reduce bleomycin-induced pulmonary fibrosis in varying degrees.3 NAC and LMWH were more obvious than Dexamethasone on the degree of improvement of pulmonary fibrosis4 IFN-γand Dexamethasone was not significantly different on the degree of improvement of pulmonary fibrosis.5 The anti-fibrosis effect of Dexamethasone,NAC,IFN-γand LMWH may resulted from expression inhibition of TGF-β1.
