【Objective】This experiment through investigating the expression and significance of matrix metalloproteinase-9(MMP-9), matrix metalloproteinase-3(MMP-3) and tissue inhibitor of metalloproteinase-2 (TIMP-2) in human lens epithelial cells (LECs) of different kinds of age -related cataract,to study the effects of MMP-9, MMP-3 and TIMP-2 and the relationship between each other in the process of age-related cataract occurrence and development.【Methods】62 cataract patients are recruited for the experiment group, among whom 23 patients (23 eyes) have posterior subcapsular cataract,24 patients (24 eyes) have cortical cataract, and 15 patients (15 eyes) have nuclear cataract;The control group have 12 normal lens. For the experiment group,we get the anterior capsule which is in the central of lens,or completely remove the lens.For the control group, completely remove the lens. We did both HE staining and immunohistochemistry to test the expression of MMP-3, MMP-9 and TIMP-2 in epithelial cells on cataract lens. Immunohistochemistry takes cells which have Buffy particles in cytoplasm or nuclear as positive, otherwise negative. Analysis of results:randomly select 5 high power views on each section and calculate positive cells percentage in the total cell number. Negative (-):positive cell percentage 0%. Weak positive (+):positive cell percentage 1%~25%. Positive (++):positive cell percentage 26~50%. Significant positive (+++): positive cell percentage more than 50%. Chi-square test and Spearman rank correlation test under SPSS software.【Results】 1:Expression of MMP-9 and MMP-3 in subcapsular cataract lens epithelial cells and cortical cataract lens epithelial cells is higher than that in nuclear cataract lens epithelial cells and normal lens epithelial cells, which is statistically significant (P< 0.05); Expression of MMP-9 and MMP-3 in cortical cataract lens epithelial cells is higher than that subcapsular cataract lens, which is statistically significant (P<0.05); The MMP-9 and MMP-3 expression difference in epithelial cells between nuclear cataract lens and normal lens is not statistically significant (P>0.05);2:The expression of TIMP-2 is no statistically significant differences between different kinds of age-related cataract lens epithelial cells(P>0.05).3:The expression of MMP-3 was positively correlated with that of MMP-9 in subcapsular cataract LECs (r=0.492, P=0.017); The expression of MMP-3 and MMP-9 was uncorrelated with that of TIMP-2 in subcapsular cataract LECs (r=0.231,-0.134; P=0.290,0.543); The expression of MMP-3 was positively correlated with that of MMP-9 in cortical cataract LECs (r=0.537, P=0.007); The expression of MMP-3 and MMP-9 was uncorrelated with that of TIMP-2 in cortical cataract LECs (r=0.184,0.301; P=0.390,0.153);【Conclusion】:1:The expression increase of MMP-3 and MMP-9 may play a important rola during the occurrence and development of cortical cataract and subcapsular cataract.2:The unbalanced expression of MMP-3/TIMP-2 and MMP-9/TIMP-2 may play a critical role during the occurrence and development of cortical cataract and subcapsular cataract.3:MMP-3 and MMP-9 may not participate in the occurrence and development of nuclear cataract.
