Atherosclerosis (atherosclerosis, AS) can lead to cardiovascular, cerebrovascular diseases, so it is serious disease threaten to human health. With the development of society and increasing competition of stress. It impact human health more and more. Therefore, the establishment of an ideal model of induction of oral tolerance on anti-atherosclerosis is significant. This study is based on establishment of atherosclerosis model, through analysed of the relationship between the oral HSP60 and AS, it can provide for the new theory and new target in cardiovascular disease.Objective:Induction of oral tolerance to HSP60 on atherosclerotic of influence and related factors.Methods:8 weeks old male ApoE-/- mice were randomly divided into high fat control group, HSP60 group. Fasted mice were gavaged with distilled water, long-chain triglycerides plus HSP60 and only distilled water separately for Every a day, totally 4 times, Abdominal injection HSP60 after 7 days from last gavage。and then a high cholesterol diet was started 24 hours after the last treatment for 10 weeks. While measuring body weight of the experiment mice every 7-10 days during the experiment. Mice were disseeted after general anesthesiato, colleeting aortic arch histopathology and producing HE stain specimen, in order to oberve the changes of aortic arch.Results:(1)After 4 weeks of high cholesterol diet:the mice in high fat control group perfomed behavior of drowsiness, eat more food, drink more water and lazy, in contrast to the HSP60 group. (2) After 4 weeks of high cholesterol diet:Bodyweight of mice were inereased in model group and control group. there was differences in weight compared with the control group was statistically significant(P<0.01), weight of model group increased more slowly than that of the control group. (3) aortic arch HE stain specimen:the plaque of aortic arch is obvious in control group in contrast to the model group, inner membrance of aortic arch is not smooth and do not see any plaque. (4) the area of plaque of aortic arch: calculate the area of plaque by digital pathological image analysis. There was differences in area compared with the control group was statistically significant (P<0.01), the area of model group (no plaque) reduced more than that of the control group (2202.89±524.25μm2).Conclusion:(1) In this study we established atherosclerotic Model by high cholesterol diet. (2) we established anti-atherosclerotic Model by oral tolerance to HSP60. (3) The animal model of this experimental can be established for general atherosclerosis by oral tolerance, which lays a foundation for the mechanism and function of oral tolerance can upgrade the level of CD4+CD25+ regulatory T cells that can guard against atherosclerosis and delay the coronary heart disease development.
