The pathological basis of coronary heart disease is coronary atherosclerosis。Atherosclerosis is a chronic progressive disease, after lipid streaks, fibrous plaque, atherosclerotic plaque of three periods. In the plaque surface may be secondary to plaque rupture, hemorrhage, thrombosis resulting in vascular dysfunction, the occurrence of cardiovascular events. Atherosclerotic process in a variety of cytokines, changes in inflammatory cytokines, immune regulation and in vivo T cell lymphocyte percentage changes have prompted the process of atherosclerosis the importance of the immune response. Through the mechanism of oral tolerance and reduce the reactivity of the immune response, and further delay the formation of atherosclerosis.HSP family is a group of widely expressed in prokaryotic and eukaryotic cells, the evolutionarily conserved protein. When not being inflammation, infection, the main role is to assist in stimulation of protein, peptide folding, assembly, play a supporting role. When stimulated, its role in the immune system may be involved in immune response, and its main role is to stimulate the body's cells macrophages, dendritic cells, natural killer cells and other innate immune cell activation, and affect the inflammatory cells Cytokines and changes in surface markers, while in the mature immune cells, differentiation, and immune signaling pathways optimization process. Taking these reasons, its more extensive study, in the HSP family, HSP60 because of its widely distributed, abundant, is stronger in more extensive clinical application.Transforming growth factor-β1 (TGF-β1), interleukin -10 (IL-10) is important in vivo inhibitor of inflammation, while involved in regulating the body's immune response involved in the occurrence of atherosclerosis development. Interferon-γ(IFN-γ) is the body's important inflammatory cytokines, the role of a wide range, can enhance the body's immune response, while activation of T cells further, B cells and promote inflammation of various parts of the release of degrading enzymes, the activation Oxygen free radicals, activation of macrophages, increased its activity, and promote further progress in inflammation. The three cytokines and atherosclerosis are closely related to the formation, so further study of its mechanism of action may further clarify the etiology of atherosclerosis, and levels of cytokines from the finding more effective treatment.Methods: 8 week old male ApoE-/- mice 16, weight 18 to 22g, were purchased from Experimental Animal Center of Peking University,and randomly divided into a atherosclerosis group (8) :high-fat diet + saline; b oral immune tolerance group (8): feeding high-fat diet + HSP60. They were given the diet for 12 weeks, cervical amputation sudden death in mice, removing from the aortic root at the aortic arch 0.5cm, placed in 4% formaldehyde solution preserved, paraffin embedded, using immunohistochemical methods, measurement and arterial plaque interleukin -10 (IL-10), transforming growth factor-β1 (TGF-β1 ), interferon-γ(IFN-γ) content, measured by the application of statistical software SPSS12.0 gray value differences between the groups compared.Results: 1) HSP60 can induce oral immune tolerance; 2) HSP60 group and high fat diet group of mice could be detected in the arterial wall of transforming growth factor-β1 (TGF-β1), interleukin -10 (IL-10), interferon-γ(IFN-γ) positive expression ;3)atherosclerosis group of mice in the aortic wall of interleukin -10 (IL-10), transforming growth factor-β1 (TGF-β1), interferon-γ(IFN-γ) levels higher than the oral immune tolerance group, and were significantly different (P <0.05).Conclusion: 1) HSP60 can induce oral immune tolerance; 2) IL-10, TGF-β1, IFN-γplay an important role in the process of atherosclerosis;3)In the process of atherosclerosis ,immunological mechanisms are involved.
