Objective:To explore the antitumor effect of different radiation pattern and investigate it of C225 combining different radiation pattern on human nasopharyngeal carcinoma CNE-2 xenografts in nude mice. And detect the expressions of Cell apoptosis genes:Bax,Bcl-2. Methods:CNE-2 cells were injected into the 48 nude mice, which were 18-20g. When the tumor grew to 0.8-1.0cm in diameter, the mice were divided into 6 arms randomly:control arm,acute irradiation model arm,IMRT arm,C225 arm,acute irradiation model+C225 arm,IMRT+C225 arm. Every arm has 8 nude mice. Explore the antitumor effect, and detect mRNA expressions of cell apoptosis genes:Bax, Bcl-2. The C225 arm, receive C225 interaperitoneal injection at the dose of 1 mg per mouse.Acute irradiation model receives 20Gy 6-MV X-ray with dose ratio 300cGy per min, while the IMRT arm and IMRT+C225 arm receive 20Gy 6-MV X-ray in 30 minutes with 15 fractions. A cute irradiation model+C225arm,IMRT+C225 arm receive C225 interaperitoneal injection at the dose of 1 mg per mouse after radiation.Then measure the maximum diameter of tumor base every other day and draw the growth curve after 40 days. And RT-PCR was used to detect mRNA expressions of cell apoptosis genes:Bax,Bcl-2. Results:1.Tumor inhibition rate:acute irradiation model, IMRT arm, acute irradiation model+C225arm and IMRT+C225 are 39.32%,27.49%,34.87% and 33.76% respectively. There is significant difference between the regular acute irradiation model and the IMRT arm (P=0.006).2. Expresssion of Bax:The expression of those arms are 0.452,0.662,0.558,0.542,0.621 and 0.555 respectively;the expression of acute irradiation model arm is higher than IMRT arm(P=0.005); Only use C225 can up the expression of Bax. But there is no significant difference between acute irradiation model arm and acute irradiation model+C225 arm, IMRT arm and IMRT+C225 arm.3. Expresssion of Bcl-2: The expression of those arm are 0.629,0.685,0.654,0.625,0.658 and 0.648 respectively. There is no significant difference between every two arms.(P>0.05). Conclusion:1. The antitumor acute irradiation model arm is better than IMRT arm. Only use C225 has no antitumor effect.。2. IMRT model prolonged fraction delivery time,and downed the expression of Bax.Only use C225 have some effect of inducing apotosis,but combine radiotheraphy have no change than only radiotherapy.3. Wether radiotherapy or combine C225 will not change the expression of Bcl-2.
